Title: Effects of different stimulation parameters of “Three Methods and Three Points” on sensory function in rats with sciatic nerve injury

Abstract: Objective: Explore the effects of strength, time, frequency on sensory function recovery in rats with sciatic nerve injury from the perspective of behavior. Methods: SD rats were used as experimental animal to make the sciatic nerve injury models, screening the appropriate parameters of “Three Methods and Three Points” which can promote the recovery of sensory function in sciatic nerve injury of rats by orthogonal test design scheme with three factors and three levels. Results: From the perspective of heat tolerance, 7 d after modeling, there was significant difference between model group and normal group (P<0.05). After 20 times of tuina intervention, tuina group and model group were significantly different (P<0.05). The best result of three factors is 2 N, 5 min, and 100/min. The primary and secondary relationship of three factors is strength>frequency>time. Conclusion: “Three Methods and Three Points” can promote the recovery of sensory function in sciatic nerve injury of rats. The most significant influence factors on recovery of sensory function is strength. Mild force, long time and high frequency stimulation are more conducive to the recovery of sensory function.

Title: Effect of acupuncture on regulating Fu organs on expression of motilin in anxiety model rats

Abstract: Objective: Effects of acupuncturing “Guanyuan” and “Xiajuxu” on the levels of motilin (MTL) and its receptors (MTLR) in both amygdala and duodenum of rats with anxiety were investigated to explore the mechanism of anti anxiety effect of acupuncture on regulating Fu organs. Methods: Clean and male SD rats were randomly divided into control group, model group and acupuncture group after 1 weeks of domestication. In both model group and acupuncture group, anxiety model was established by chronic unpredictable stress (CUS) method. Rats in acupuncture group were treated with “Guanyuan” and “Xiajuxu”. The anxiety behavior of rats was observed by elevated plus maze (EPM) (OT% and OE% represented the percentage of time and frequency of the open arm occupying total time and total number of times of the rats in the cross maze respectively). The concentrations of MTL and MTLR in the brain and intestine and the content of plasma corticosterone (CORT) of rats were detected by enzyme-linked immunosorbent assay (ELISA) method. Results: 1) Compared with control group, OT% of model group was decreased significantly (P<0.05) and OE% was also decreased (P=0.051). Compared with model group, OT% of acupuncture group was increased significantly (P<0.05) and OE% was also increased (P=0.054). 2) Compared with control group, concentrations of MTL in the amygdala and MTLR in the duodenum of model group were decreased (P=0.067 and P=0.078). Compared with model group, concentrations of MTL in amygdala and MTLR in duodenum of acupuncture group were increased, but there was no significant difference among them (P>0.05). 3) Compared with model group, the content of plasma CORT of acupuncture group was decreased (P=0.065). Conclusion: Anti anxiety effect of acupuncture on regulating Fu organs may be related to up-regulating the concentrations of MTL in amygdala and MTLR in duodenum and down-regulating activity of hypothalamic-pituitary-adrenal (HPA) axis.

Title: Effects of Chaiqi decoction on oxidative stress in the liver tissue from NAFLD rats

Abstract: Objective: To observe the effects of Chaiqi decoction on oxidative stress in the non-alcoholic fatty liver disease (NAFLD) rat model, and analyze its protection for liver tissue. Methods: 32 rats were randomly divided into normal group, model group, Chaiqi decoction group and rosiglitazone group. The rat model was established by feeding high fat, high sugar and high salt diet for 8 weeks. The Chaiqi decoction group was given Chaiqi decoction (5.67 g•kg-1•d-1) from the beginning of modeling, and rosiglitazone group was given rosiglitazone (3 mg•kg-1•d-1). The levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) in serum and the levels of superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GSH-Px), triglyceride (TG) in liver tissue were detected after 8 weeks. Liver pathological histopathology among the 4 groups were observed under optical microscope. Results: In comparing with normal group, the levels of ALT, AST in serum and the levels of MDA, TG in liver tissue were significantly increased, while the levels of SOD and GSH-Px in liver tissue were decreased in model group (P<0.05). All the indexes of the two treatment groups were lower than model group, the difference was statistically significant (P<0.05). There was no significant difference between Chaiqi decoction group and rosiglitazone group in the levels of ALT and AST (P>0.05), but the levels of SOD and GSH-Px in liver tissue in Chaiqi decoction group were higher than those of rosiglitazone group. The difference was statistically significant (P<0.05). Conclusion: Chaiqi decoction and rosiglitazone both have positive therapeutic effects against hyperlipidemia and abnormal liver function in experimental NAFLD rats, better than rosiglitazone in oxidative stress.

Title: Simultaneous determination of berberine hydrochloride, baicalin and emodin in Sanhuang tablets by HPLC

Abstract: Objective: A rapid and specific high performance liquid chromatography (HPLC) method was developed for the analysis of berberine hydrochloride, baicalin and emodin. The method was applied to study the content determination of Sanhuang tablets containing berberine hydrochloride, baicalin and emodin. Methods: The analysis was carried out on a Agilent C18 reversed-phase column (4.6 mm × 250 mm, 5 μm) by gradient elution with acetonitrile and 0.5% phosphoric acid (adjusted pH=4.0 with triethylamine). The flow rate was 1 mL•min-1 and the detection wavelength was set at 275 nm. Results: Linear responses were obtained for berberine hydrochloride and baicalin ranging from 1 to 30 μg∙mL-1, emodin ranging from 0.5 to 15 μg∙mL-1. The extraction recovery ranged from 95.83% to 100.09%. Conclusion: This method is simple, accurate with good repeatability, low detection limit and can eliminate the interference of other components. This validated method could be used in the quality control of Sanhuang tablets.

Title: Study on the synthesis of polysubstituted tetrahydro-β-carboline derivatives

Abstract: The β-carboline alkaloid is an important type of indole alkaloids, and has attracted increasing interest of synthetic chemists, owing to its wide distribution in the nature as well as various significant biological activities. Herein, we report a synthesis study of chiral 2, 3, 9-trisubstituted-tetrahydro-β-carboline derivatives with commercially available L-tryptophan as the starting material, with Pictet-Spengler reaction and Stille coupling as the key steps. This study paves the base for the further biological studies of β-carboline alkaloid derivatives.

Title: Synthesis of α7-nAChR agonists drugs and druggability evaluation

Abstract: Objective: To study the synthesis routes of α7-nAChR agonists and evaluate the toxicity and pharmacokinetics of N-[(3R)-1-azabicyalo [2.2.2] octane-3-yl]-5-phenyl-indolizine-2-carboxamide (compound X1). Methods: Compound X1 was synthesized with Suziki, Baylis-Hillman, thermocyclic reaction, et al. Evaluation on druggability mainly included acute toxicity test, guinea-pigs cardio-toxicity test and pharmacokinetic study. Results: Acute toxicity tests showed that median lethal dose (LD50) of compound X1 was more than 2 000 mg/kg, while that of EVP-6124 was 1 882.7 mg/kg, both blonging to low toxicity compounds. Guinea-pigs cardio-toxicity test illuminated that compound X1 appeared cardio-toxicity. When dose of compound X1 was 10, 20 mg/kg , the QT and QTc in anesthetized guinea-pigs were prolonged by more than 10%. While, EVP-6124 with 10 mg/kg still didn’t occur significant prolongation in QT. Absorption and excretion of compound X1 was slow, of which the bioavailability was 53.2%. Compound X1 could pass the blood brain barrier and accumulated in brain tissue. Conclusion: Compound X1 can be a candidate with low acute toxicity and good pharmacokinetic properties. The study provides significant preclinical data for schizophrenia.

Title: Quantitative detection of Salmonella by dual recognition based on Man-Fe3O4 and Man-CQDs nano-probes

Abstract: In the paper, citric acid was used as carbon source for preparing fluorescent carbon quantum dots (CQDs) by hydrothermal method. Due to the obtained CQDs rich in carboxyl group on the surface, Man-CQDs nano-probe was synthesized by immobilizing 4-aminopheny-α-D-mannopyranoside on surface of CQDs via amido bond. Man-Fe3O4 nano-probe was obtained by the same way on the commercial Fe3O4. In the mixed bacteria solution, Salmonella was selectively captured by Man-Fe3O4 nano-probe, and Bacteria-Man-Fe3O4 complexes were isolated by exerting external magnetic field. Moreover, the captured Salmonella was labeled by Man-CQDs. Salmonella labeled dual recognition was tested by fluorescence spectroscopy. The results show that there is a good linear relationship between the logarithm of the concentration of Salmonella and the corresponding fluorescence value. The linear equation is IFL=217.2log(c)-758.8 (R2=0.957 8), with the linear range of 104-107 cfu•mL-1 and the detection limit of 5×103 cfu•mL-1. A method for detection of Salmonella based on the magnetic nano-probes is developed, which has the advantages of fast, high selectivity and high sensitivity.

Title: Investigation of Tat impairing anti-Cryptosporidium parvum defense mechanism

Abstract: The purposes of this paper are to investigate the effects of HIV Tat protein on miRNA-mediated epithelial anti-Cryptosporidium parvum defense. Using quantitative real-time PCR (qRT-PCR), Western blotting and luciferase assay, we demonstrated that HIV Tat decreased the expression of inducible nitric oxide synthase (iNOS) and induced epithelial expression of KH-type splicing regulatory protein (KSRP) (also known as KHSRP) through down regulation of selected miRNAs, such as miR-27b. As an RNA-binding protein, KSRP could recognize AU-rich element (ARE) within the 3'-untranslated region (3'-UTR) of iNOS mRNA and control their half-life time in the cytoplasm which impairs the defense of epithelial cell to C. parvum infection. Results from our studies could provide a rational basis for the implementation of new immunotherapeutic strategies for opportunistic infections in AIDS patients.

Title: Purification and identification of EPCs derived from peripheral blood

Abstract: Objective: The endothelial progenitor cells (EPCs) derived from peripheral blood with excellent ability of vascularization were isolated and purified, to supply high quality and easily obtained seed cells for tissue engineering vascularization. Methods: EPCs were isolated from rabbits peripheral blood with density gradient centrifugation and cell attachment, and cultured in endothelial cell medium (ECM). The expressions of CD34, CD133 and vWF on the third generation adherent cells were detected with immunofluorescent staining, and EPCs were identified with immunohistochemical double staining. Results: Higher purity EPCs could be isolated with Ficoll density gradient centrifugation and cell attachment methods. The CD34, CD133 and vWF of the third generation peripheral blood derived EPCs were positive with immunofluorescent staining, and DiI labeled acetyl low-density lipoprotein cholesterol and fluorescein labeled lectin immunofluorescence double staining were positive, which were consistent with the EPCs characteristic. Conclusion: EPCs isolated from rabbits peripheral blood with Ficoll density gradient centrifugation and cell attachment were highly purified.

Title: A staining method to detect cellular culture state on hydrophilic scaffold

Abstract: Objective: In present study, a method was introduced to detect the state of cells cultured on the hydrophilic scaffold. Methods: Stem cells were cultured on a hydrophilic scaffold to build a cell-scaffold complex with a confluence of 90%-95%. The fluorescence staining of cell-scaffold complex was produced by acridine orange staining, and rinsed by PBS to erase the overdoes acridine orange dyes. After fluorescent staining, the cell-scaffold complex was observed under green and orange channels of laser scanning confocal microscope. Results: By this method, the state and distribution of cultured cells could be displayed clearly and precisely under laser scanning confocal microscope, without the interference of unintended staining on hydrophilic scaffold. Conclusion: The present study provides a harmless and simple method to detect the distributions and growth states of cells cultured on the hydrophilic scaffold, and it is suitable for dynamic monitoring of the states of cultured cells, and useful for wild-spread.

Title: Protective effects of AS-IV on high insulin-induced human mesangial cell injury and its mechanisms study

Abstract: Objective: To research the protective effects of astragalosides IV (AS-IV) on high insulin (HINS)-induced human mesangial cell (HMC) injury in vitro and its potential mechanism. Methods: HMC was taken as target cell. The HMCs were divided and treated as follows: 1) normal group (NG), insulin group (INS, respectively treated with 2, 4, 8, 16, 32, 64, 128 nmol/L insulin); 2) normal group (NG), high insulin group (HINS, 128 nmol/L), AS-IV group (25, 50, 100 μmol/L), Tempol group (100 μmol/L), LY294002 group (10 μmol/L). After treating for 12 h, the proliferation and viability were analyzed by MTT assay. The expression of reactive oxygen species (ROS) produced in HMC was measured by DCFH-DA method. Western blotting was used to test the expression of total Akt, p-Akt, total gsk-3β, p-gsk-3β and TRPC6 proteins. Flu-3/AM was used as a fluorescent probe for detecting intracellular calcium ion concentration in each group. Results: The proliferation of HMC was enhanced in dose- and time-dependent manners by increasing concentrations and time of insulin. AS-IV inhibited proliferation of HINS-induced HMC in a dose-dependent manner compared to HINS group, AS-IV treatments significantly inhibited HMC proliferation, reduced intracellular ROS contents and decreased the expression of p-Akt and p-gsk-3β, increased the expression of TRPC6 and intracellular calcium ion concentration. Conclusion: AS-IV has protective effects on HMC injury induced by HINS, which relate to increasing antioxidant ability of HMC, reducing p-Akt, p-gsk-3β protein contents, increasing expression level of TRPC6 and intracellular calcium ion concentration.

Title: Study on the expression of Pak2 in chemotherapy resistant ovarian serous cancer tissues and cells and its clinical significance

Abstract: Objective: To detect the differential expression of p21 protein activated kinase 2 (Pak2) in chemotherapy resistant and sensitive ovarian serous cancer tissues and cells. Methods: Immunohistochemistry analysis and Western blotting were applied for the detection of Pak2 in chemotherapy resistant and sensitive ovarian serous cancer tissues. Using quantitative real-time PCR (qRT-PCR) method to detect the differential expression of Pak2 in serous ovarian cancer resistant SKOV3-TR30 and sensitive SKOV3 cells. Results: Immunohistochemistry and Western blotting assay showed that the expression of Pak2 in chemotherapy resistant ovarian serous cancer tissues was significantly higher than the sensitive tissue, and mainly in the cytoplasm. By applying qRT-PCR and Western blotting, we found that there was no significant difference of Pak2 mRNA expression levels between these two cell lines. The expression of Pak2 protein was significantly over-expressed in ovarian cancer paclitaxel resistance cell line SKOV3-TR30 than in its parental cell line SKOV3. Conculsion: Pak2 expression in chemotherapy resistant ovarian serous cancer tissues and cells was significantly higher and the over expression was associated with drug resistance.

Title: Role of aberrantly glycosylated IgA1 and miRNAs in pathogenesis of IgA nephropathy

Abstract: IgA nephropathy (IgAN) is characterized by the diffuse deposition of IgA1 in mesangial area of glomerulus. IgAN is the most common primary glomerular disease, and also the major cause of end-stage renal disease (ESRD) in China. In recent years, great progress has been made in the research of galactose-deficient IgA1 (Gd-IgA1) and miRNAs in IgAN, which significantly improves our understanding on the pathogenesis of IgAN. In this paper, the progress of Gd-IgA1 and miRNAs in IgAN is reviewed, which establishes a theoretical foundation for future clinical treatment.

Title: Influence of career growth expectation of new medical staffs in township hospital on early turnover intention

Abstract: Objective: To introduce the concept of “career growth expectation” into the groups of new medical staffs in township hospital, and to explore the influence of career growth expectation on early turnover intention. Methods: Using an online questionnaire survey to collect 263 valid questionnaires, exploratory factor analysis and multiple linear regression analysis were used for data analysis. Results: Career growth expectation scale included 14 measurement items under 2 dimensions. The internal consistency coefficient of scale was 0.920. Career growth expectation scale had a good reliability and predictive validity. Career growth expectation of new medical staffs in township hospital had a significantly negative prediction (β=-0.289, P<0.01) on early turnover intention. Conclusion: Career growth expectation scale has a good reliability and validity among new medical staffs in township hospital, meanwhile, with a high level of career growth expectation of new medical staffs, they have a strong sense of belonging and person-organization fits, which will reduce the early turnover intentions of staffs.

Title: Study on reproductive toxicity of arsenic

Abstract: Arsenic-induced reproductive system toxicity has attracted more and more attention. The study of epidemiological data and animal experiments have confirmed the effect of arsenic reproductive toxicity on exposure persons. This paper summarizes the arsenic-induced human and animal reproductive toxicity studies in recent years from the aspects of changes in germ cells, embryos and hormones, including decreased sperm counts and viability, increased sperm deformity, impaired testicular tissue structure, changed testosterone and luteinizing hormone levels, and miscarriage, deformity, stillbirth and low birth weight and other reproductive effects of children caused by maternal arsenic exposure, laying a theoretical foundation for the future study of arsenic reproductive toxicity.
