Abstract:It has been proven that epigenetic mechanisms such as histone post-translational modification via altering the chromatin structure and heritably regulating the gene expression are involved in most cellular biological processes. Thus, abnormal regulation of epigenetics is implicated in the occurrence of various diseases, includong cancer. Human MYST1 (also known as hMOF, KAT8) is a member of the MYST (Moz-Ybf2/ Sas3-Sas2-Tip60) family of histone acetyltransferases (HATs). As a catalytic subunit, hMOF can at least form two distinct multiprotein complexes (MSL and NSL) in human cells. Both complexes can acetylate histone H4 at lysine 16 (H4K16), however, NSL complex possesses broader substrate specificity and can also acetylate histone H4 at lysine 5 and 8 (H4K5 and H4K8), suggesting the complexity of intracellular functions of hMOF. Silencing of hMOF in cells leads to genomic instability, reduced transcription of certain genes, defective DNA damage repair and early embryonic lethality. Abnormal gene expression of hMOF and its corresponding acetylation of H4K16 have been found in certain primary cancer tissues including breast cancer, medulloblastoma, ovarian cancer, renal cell carcinoma, colorectal carcinoma, gastric cancer as well as non-small cell lung cancer. In this review, we provide a brief overview of the hMOF and its corresponding histone acetylation, introduce recent research findings that link hMOF functions to tumorigenesis, and speculate on the potential role that may be relevant to tumorigenic pathways.

	Abstract:BRCA2- and CDKN1A-interacting protein (BCCIP) has been considered an important cofactor for BRCA2 in tumor suppression. We previously observed the down-regulation of BCCIP in multiple clinically diagnosed primary tumor tissues including ovarian cancer, renal cell carcinoma and colorectal carcinoma. In this study, we first demonstrate that BCCIP expression was regulated by human INO80 (hINO80) chromatin remodeling complex in cells. In chromatin immunoprecipitation (ChIP) assays, we show that the catalytic subunit hINO80 protein and YY1, a subunit of hINO80 complex, co-localize the BCCIP promoter region (+0.25 kb downstream of the BCCIP), and the existence of hINO80 is required for recruiting the hINO80 complex to BCCIP promoter. RNAi knockdown of hINO80, Arp8 or YY1 resulted in reduction of BCCIP expression both in mRNA and protein level. In contrast, over-expression of hINO80, Arp8 or Arp5 enhanced the luciferase activity of pGL4-BCCIP. Together, as a novel target gene, BCCIP is regulated by hINO80 chromatin remodeling complex. Our findings will provide: 1) a theoretical basis to further elucidate the collaborative functions between the hINO80 complex and BCCIP in human cells; 2) an experimental basis for clarifying the role of BCCIP and BRCA2 in tumor suppression. ?????

	Abstract:Glioma stem cells (GSCs) are a subset of tumor cells that drive glioma initiation and progression. The molecular mechanisms underlying the maintenance of GSC are still poorly understood. Here we investigated the role of RhoGDIα in GSCs. Over-expression of RhoGDIα suppressed the self-renewal and promoted the differentiation of GSCs. Further data suggested RhoGDIα inhibited the transcription of Oct4 through RhoA/ROCK1/FOXO1 pathway. Moreover, inactivation of ROCK1 also decreased the self-renewal and Oct4 transcriptional activity, and rescued the effects caused by RhoGDIα knockdown. Our results indicate that RhoGDIα is involved in the maintenance of GSCs.

	Abstract:To comprehend the gene expression profile in rice panicle under high temperature, Agilent 4×44K rice oligo microarray experiments were carried out using post-meiosis stage rice panicle treated at 40 centigrade degree for 0 min, 10 min, 20 min, 60 min, and 2 hr. The time course differentially expressed genes under heat stress were mainly involved in binding, catalysis, stress response, and cellular process. The significantly changed genes during heat treatment were mainly up-regulated. Among heat-responsive genes, the predominant transcription factor gene families were Hsf, NAC, AP2/ERF, WRKY, MYB, and C2H2. The MapMan analysis demonstrated that, under heat treatment, the HR genes were enriched in the pathways related to biotic and abiotic stress, cell cycle, development, ubiquitin-proteasome system, lipid and secondary metabolisms. These data revealed the great importance of cross-talk and protein homeostasis in response to heat stress in rice panicle at post-meiosis stage.

	Abstract:In this paper………..(10 Points, Times New Roman) WRKY proteins are a large family of regulators involved in various developmental and physiological processes, especially in coping with diverse biotic and abiotic stresses. In this study, a comprehensive analysis of the Populus genomic sequences showed that the structures of 100 predicted PtrWRKY genes contained complete WKRY domain. Phylogenetic analysis revealed that the members of this superfamily among poplar, Arabidopsis and other species were divided into three groups with several subgroups based on the structures of the WRKY conserved domains. Various cis-acting elements related to stress and defense responses were found in the promoter regions of PtrWRKY genes by promoter predicted analysis. High-throughput transcriptomic analyses identified that 61 of the PtrWRKY genes were induced by biotic and abiotic treatments, such as Marssonina brunnea, salicylic acid (SA), methyl jasmonate (MeJA), wounding, cold and salinity. Among these PtrWRKY genes, transcripts of 41 selected genes were observed in different tissues, including roots, shoots and leaves. QRT-PCR analysis further confirmed the induced expression of 19 PtrWRKY genes by one or more stress treatments. The overexpression of a SA-inducible gene, PtrWRKY89, accelerated expression of PR protein genes and improved the resistance to pathogens in transgenic poplar, suggesting that PtrWRKY89 was a regulator of SA-dependent defense signaling pathway in poplar. Taken together, our results provided signi?cant information for improving the resistance and stress tolerance of woody plants.

	Abstract:Hsp90 is vitally important among the various HSPs because its target proteins are always steroid hormone receptors, signaling kinases, and a variety of transcription factors involved in various tumors. However, the mechanism by which Hsp90 regulates the crosstalk between 20-hydroxyecdysone (20E) and juvenile hormone (JH) signal pathway is not clear. Alignment analysis shows that H. armigera Hsp90 is highly conserved across invertebrates and vertebrates. Western blots indicate that Hsp90 expression is regulated during insect development. The Hsp90 inhibitor 17-N-allylamino-17-demethoxygeldanamycin represses the 20E- or JH-induced gene expression. The results suggest that Hsp90 may participate in the 20E and JH signal pathway via regulating the 20E- and JH-inducible gene expression.

	Abstract:Reducing power is an essential factor for fatty acid biosynthesis. In this study, we investigated the effect of five reducing power generating enzymes (NAD-dependent malic enzyme (NAD-ME), NADP-dependent malic enzyme (NADP-ME), NADP-dependent isocitrate dehydrogenase (NADP-IDH), glucose 6-phosphate dehydrogenase (G6PD) and 6-phosphogluconate dehydrogenase (PGD)) on fatty acid biosynthesis in an engineered E. coli BL21 △fadE/pTE which has a fatty acid metabolic sink. Overexpression of NAD-ME, NADP-IDH and G6PD resulted in 2.15-, 1.6- and 1.16-fold increase in fatty acid yield respectively, while overexpression of PGD didn't lead to a significant increase of fatty acid production and overexpression of NADP-ME led to a 15% decline in fatty acid yield. This study provides a feasible strategy for enhancing fatty acid production in engineered E. coli strain by overexpressing the reducing power generating enzymes NAD-ME and G6PD.

	Abstract:Drosha/DGCR8 complex cleaves primary microRNAs (pri-miRNAs) to yield ~ 65 nucleotide (nt) precursor microRNAs (pre-miRNAs). Dicer cleaves pre-miRNAs to yield ~ 22 nt miRNA/miRNA* duplexes. Interactions of Drosha/DGCR8 and Dicer with their substrates are incompletely understood. By in vitro Drosha and Dicer assays, we proposed models that human Drosha/DGCR8 and Dicer recognize their substrates in different manners. The flanking sequences and the terminal loop structure of a pri-miRNA are critical for Drosha/DGCR8 processing pri-miRNAs. The terminal loop of a pre-miRNA is not essential for hDicer processing. The end side and the stem of the pre-miRNA contribute to the recognizing and binding with Dicer. This study improved our understanding on RNase III functional mechanism and miRNA processing.

	Abstract: It is very certain that each on of us will meet death one day, but people are trying every way to live longer. So age-related study is consequently hot nowadays. As ageing study has became increasingly popular recent days. This meta-analysis is aiming at showing the latest progress on ageing study published on NATURE from Jan 2011 to Dec 2012 and analysis the scientific methods they used in these researches, and the data obtained from the scientific methods used in research through statistics methods can provide with references to those who are finding proper methods for their own researches. The analysis will present in mainly two terms, on recent findings and on research methods analysis. Researches on human cognitive decline with age and functional gene analysis are the main subjects of findings. Analysis on research methods has revealed that SNP related method is frequent used in ageing related statistical study as well as ageing related cognition study. And about 2/3 of the researches are conducted with the aid of molecular biology methods.

	Abstract:One factor involved in eukaryotic translation termination is Class 1 release factor in eukaryotes (eRF1), which functions to decode stop codons. Variant code species, such as ciliates, frequently exhibit altered stop codon recognition. Studies revealed that some class-specific residues in the eRF1 N-terminal domain are responsible for stop codon reassignment in ciliates. Here, we investigated the effects on stop codon recognition of chimeric eRF1s containing the N-terminal domain of Euplotes octocarinatus and Blepharisma japonicum eRF1 fused to Saccharomyces cerevisiae M and C domains using dual luciferase read-through assays. Mutation of class-specific residues in different eRF1 classes was also studied to identify key residues and motifs involved in stop codon decoding. As expected, our results demonstrate that three pockets within the eRF1 N-terminal domain were involved in decoding stop codon nucleotides. However, allocation of residues to each pocket was revalued. Our data suggest that hydrophobic and class-specific surface residues participate in different functions: modulation of pocket conformation and interaction with stop codon nucleotides, respectively. Residues conserved across all eRF1s determine the relative orientation of the three pockets according to stop codon nucleotides. However, quantitative analysis of variant ciliate and yeast eRF1 point mutants did not reveal any correlation between evolutionary conservation of class-specific residues and termination-related functional specificity, and was limited in elucidating a detailed mechanism for ciliate stop codon reassignment. Thus, based on isolation of suppressor tRNAs from Euplotes and Tetrahymena, we propose that stop codon reassignment in ciliates may be controlled by cooperation between eRF1 and suppressor transfer RNAs
