Abstract:Heat shock proteins (HSPs) have been found to be ubiquitous. More than 70 putative HSP genes were found in rice (Oryza sativa L.), but few of them have been characterized. Expression patterns of nine rice OsHSP genes were analyzed by semi-quantitative RT-PCR. The nine OsHSPs were named OsHSP80.2, OsHSP74.8, OsHSP71.1, OsHSP58.7, OsHSP50.2, OsHSP26.7, OsHSP24.3, OsHSP24.1 and OsHSP16.9 respectively according to their approximate molecular weight. The nine genes showed different expression profiles in roots, leaves, leaf sheaths, spikes and embryos. All nine OsHSP genes were induced strongly by heat shock treatment, whereas none of them were induced by cold. The transcripts of OsHSP80.2, OsHSP71.1 and OsHSP24.3 were increased during salt tress treatment. Expression of OsHSP80.2 and OsHSP24.1 genes were enhanced while treated with 10% PEG. Only OsHSP71.1 was induced by ABA while OsHSP24.1 was inhibited by ABA. | |||

Abstract:Mitochondrial 12S rRNA, 16S rRNA, cytochrome b, ATP synthase 8 and 6 gene fragments of Chi-lin fish from Tai Mountain were amplified by PCR, the products were purified and sequenced. The sequences and characters of the multiple genes of mitochondrial DNA (mtDNA) were analyzed by MEGA 3.1. The results showed that all of the multiple genes were A-rich; the base compositions of the protein-coding genes were globally G-deficient, especially in the third position. Interestingly, transitions of protein-coding genes were major in the substitution, and largely concentrated in the third position, too. All molecular phylogenetic trees revealed that Chi-lin fish did not belong to genus Varicorhinus, against the former morphological data. And it was comparatively closer to genus Spinibarbus and Barbus. The Cyt b sequence analysis suggested that the divergence time of Chi-lin fish was about 16.0 Mya (million years old) when Tai Mountain was just in Middle Miocene and suffered the aftereffect of Yan Shanian movement. It inferred that the vicariance events were the primary momentum to promote forming the limited distributing of the present. | |||

Abstract:Two uridine auxotrophic mutants of Trichoderma reesei were isolated by resistance to 5-fluoroorotic acid after UV mutagenesis. One mutant, called M23, was complemented with the Aspergillus niger pyrG gene carried by plasmid pAB4-1. Mutated pyrG gene of M23 was cloned and DNA sequencing analysis indicated that a cytosine inserted into the 934-939 oligo dC position of the pyrG coding region, resulting in a frameshift mutation. Transformation efficiency was approximately 200-300 transformants per μg of DNA with plasmid pAB4-1. Stable transformants were obtained by monosporic culture and showed to be prototroph after successive propagation. Vitreoscilla hemoglobin expression plasmid pUCVHb co-transformed with plasmid pAB4-1 attained transformation efficiency of 71.8% versus 21.6% with pAN7-1. Southern blot analysis of the transformants demonstrated that plasmid pUCVHb was integrated into the chromosomal DNA. The experimental results demonstrated that the pyrG-based system was more efficient and timesaving than the conventional hygromycin B resistance-based transformation system. | |||

Abstract:In order to investigate the effect of calnexin deletion on the induction of main ER molecular chaperone BiP, we cultured the wild-type and calnexin-disrupted Saccharomyces cerevisiae strains under the normal and stress conditions. The growth rate of the calnexin-disrupted yeast was almost the same as that of the wild-type yeast under those conditions. However, the induced mRNA level of BiP in the ER was evidently higher in calnexin-disrupted S. cerevisiae relative to the wild type at 37 C, but was almost the same in the two strains under the normal conditions. The western blotting analysis for BiP protein expression in the ER yielded results that showed a parallel in their mRNA levels in both strains. It is suggested that under heat stress conditions the induction of BiP in the ER could recover parts of the function of calnexin in calnexin-disrupted yeast, and result in the same growth rate as in wild-type yeast. | |||

Abstract:The cDNAs encoding two distinct growth hormone receptors were firstly cloned and sequenced from the liver of orange-spotted grouper (Epinephelus coioides). The cDNA of grouper GHR1 consisted of 2673-bp and encoded 658 amino acids and that of grouper GHR2 consisted of 2989-bp and encoded 577 amino acids. The two cDNAs had 78.6% identity in nucleotide sequence and 37.8% identity in deduced amino acid sequence. Northern blot analyses demonstrated a single GHR1 transcript of approximately 4.3Kb in liver and a single GHR2 transcript of approximately 3.9Kb in the liver and muscle. In the Real-time PCR assay, grouper GHR1 and GHR2 were expressed in all tissues tested, and the expression of GHR2 was significantly higher than GHR1 in telencephalon, cerebellum, pituitary, heart and white muscle, whereas the expression of GHR1 was significantly higher in liver. The grouper GHR1 had high homology to somatolactin receptor (SLR) and the grouper GHR2 might play a role of modulator of grouper GHR1. These results indicated that there were two types of GHR existing in orange-spotted grouper and they had different structural features and physiological functions. However the general functional differences between GHR1 and GHR2 of orange-spotted grouper remained further research. | |||

Abstract:A lipase-producing bacterium strain B68 was screened and identified as Pseudomonas fluorescens sp. Novel lipase gene lipB68 was cloned and sequenced… by the method of GenomeWalker. Sequence analysis revealed an open reading frame of 1,431 nucleotides, encoding a protein with 476 amino acids and a molecular mass of 50,193 Da, being a member of lipase subfamily I.3. The lipB68 gene was expressed in E. coli BL21 (DE3) in the form of inclusion body and the purification and protein refolding were conducted. Lipase lipB68 had a favorable reaction temperature of 20°C with p-Nitrophenyl caprate as substrate, lower than all the known lipases from subfamily I.3. And it also showed good stability to organic solvents such as DMF and acetonitrile. After being immobilized on fabric of cellulose, lipase lipB68 showed high enantioselectivity at the resolution of racemic a-phenylethanol and a-phenylpropanol (E=220, and 190 respectively). | |||

Abstract:Two wild type strains of Acidithiobacillus ferrooxidans, which were called CMS and YTW respectively, were isolated from low-grade copper ores in China. The data of iron oxidation and growth rates shows that strain YTW was much lower iron oxidation capacity and longer generation time than strain CMS. To reveal these heterogeneous phenotypic characters, the iro gene encoding Iron oxidase, which catalyzed the first step of iron oxidation metabolism system, was amplified by PCR and sequenced for both strains. Sequence alignment showed that there was a Threonine(27) -to- Proline(27) substitution in the signal peptide of Iron oxidase for strain YTW. The protein secondary structure predicted that the substitution made a loop among the residue 26, 27 and 28. This substitution may affect the location and interaction of Iron oxidase in cellular membrane. The results implied that Proline(27) substitution in the signal peptide of Iron oxidase may be an important factor for heterogeneous iron oxidation capacity of iron oxidation metabolism system. Abstract:he present studies described characterization of two rice bentazon-lethal mutants, bel1 and bel2, which were created via γ-ray radiation in an indica photo-thermo-sensitive genic male sterile (P/TGMS) line, W6154S, and a japonica inbred line, Norin8, respectively. Both mutants of bel1 and bel2 started to show affected symptoms at a concentration of 100 mg/L and became lethal at 300 mg/L bentazon but within this range bel1 is slightly more tolerance than bel2. The lowest concentrations of their bentazon-sensitivity, however, were about 120-fold lower than those of their wild controls. Using bel1 to cross with bel2 followed by herbicide test of their progenies, we confirmed that these two mutants were genetically allele each other. The data from PCR amplification followed by sequence comparisons between wild and mutant alleles revealed a single base-pair deletion in each mutant and for bel1 this occurred at 408th bp in its second exon, while for bel2 this at 507th bp in its first exon. | |||
