Abstract:To characterize the E2 profiles after oocyte pick-up (OPU) and assess the predictive value of serum E2 in early diagnosis of pregnancy following IVF-ET, in this study sixty-two patients (75 cycles involved) undergoing IVF-ET cycles whose infertility was attributed to tuble factors or male factors were enrolled. Serum E2 was detected on alternate day after OPU with micro-particle enzyme immunoassay. It was found that the level of serum E2 declined progressively after OPU in both pregnant and non-pregnant cycles and showed no significant difference on day 2, 4, 6, 8 days after OPU. In pregnant cycles following IVF-ET, serum E2 reached nadir on day 10 and then increased gradually. The differences of serum E2 level in pregnant and non-pregnant cycles were detectable from day 10 after OPU (816.4±537.6.vs 189.5±69.3 pg/ml) (P<0.05). In non-pregnant cycles, day 10 E2 in was significantly lower than that of day 8(189.5±69.3 vs 989.2±581.5 pg/ml) (P<0.05). We concluded that assessing E2 concentration on day 8 and 10 consecutively after OPU may have predictive value in diagnosing early pregnancy. When prominent drop appears, it’s very likely to see implantation failure. | |||

Abstract:Objective: To investigate the curative effect of emergency stent placement for left main bronchial malacia, stenosis and collapse after orthotopic heart allotransplantation. Methods: To evaluate the risk and efficacy of treatment of bronchus stent placement after orthotopic heart allotransplantation in one dilated cardiomyopathy patient complicated with left main bronchial malacia, stenosis and collapse resulting in left pulmonary ventilation dysfunction, and other multi-diseases, such as severe pulmonary artery hypertension and mixed(mainly central) sleep apnea syndrome. Results: After treatment of stent placement, patient was relieved from left pulmonary ventilation dysfunction with obviously improved hypercapnia, hypoxemia, pulmonary hypertension and the function of transplanting heart recovered. Conclusions: Emergency treatment with stent placement for bronchial malacia, stenosis and collapse occurring after orthotopic heart allotransplantation could improve ventilation dysfunction caused by bronchial malacia and stenosis, and increase the survival rate of heart transplantation. | |||

Abstract:The promising results in replacement of the beta-cell mass have been accomplished by beta-cells implanting. However, the demand for beta-cells is far greater than the supply. The transdifferentiation of stem cells into insulin-secreting cells could potentially provide a new help for beta-cell supplying. To determine whether the myoblast cell line C2C12 could be induced to transdifferentiate into insulin-secreting cells by pancreatic-duodenum homeobox-1 (PDX-1). A myoblast cell line (C2C12) was transfected by the PDX-1 constructing to overexpress PDX-1 protein and then treated with various concentrations of glucagon-like peptide-1 (GLP-1). The Expression of PDX-1 and insulin mRNA was determined by in situ hybridization and the intracellular insulin was determined by immunocytochemistry. The result shows that the PDX-1 transfected cells expressed more insulin and secreted more insulin than the vector transfected cells. We conclude that PDX-1 and GLP-1 might induce myoblasts to transdifferentiate into insulin-secreting cells. | |||

Abstract:nvolvement of AFP against apoptosis of tumor cell has been implicated in its evasion of immune surveillance. However, the molecular events of immune escape mechanisms are still unknown. The major observations reported here relate to a possible mechanism by which heptoloma Bel 7402 cells escape immune surveillance in vitro. Western blotting and a well-characterized cofocal scanning image were performed to analyze the expression of Fas/FasL and caspase-3 in co-cultured Bel 7402 and Jurkat cells. After co-culture with Jurkat cells, up-regulated Fas and reduced FasL expression could be observed. Treatment with AFP could remarkably inhibit the elevated Fas and, whereas, induce the FasL expression in co-cultured Bel 7402 cells. Cells co-culture could induce the expression of caspase-3 in both cells line. The elevated caspase-3 in Bel 7402 cells was abolished following the treatment of AFP. The expression of caspase-3 was elevated in co-cultured Jurkat cells treated with AFP. No detectable | |||
