Abstract:Malignant fibrous histiocytoma (MFH) is a malignant tumor composed of a mixture of fibroblastic and histiocytic cells. A rare case of primary MFH of the breast was reported and thirty-eight Chinese cases in recent twenty-two years were reviewed. Their clinical features and survival information were described and compared with those in English literatures. The 5-year survival rate of the Chinese MFH patients was 57.1%. Large tumor and skin invasion were related with bad prognosis. Simple mastectomy is recommended especially for orient patients. | |||

Abstract:Introduction: Breast cancer lacks good serum tumor markers for its diagnosis, follow-up and evaluation of curative effect. Our research aimed to use SELDI-TOF-MS and CM10 ProteinChip to detect the serum proteomic patterns in patients of breast cancer, screen the biomarker candidates, build and validate the diagnostic models and evaluate its clinical value in surveillance and follow-up after operation. Methods: SELDI-TOF-MS technology and CM10 ProteinChip were used to detect the serum proteomic patterns of 63 breast cancer patients and 40 healthy women. ZJU-PDAS software was used to screen the biomarker candidates and build the diagnostic models. The best model was blind validated in the subsequent-recruited 23 patients and 20 healthy women. Another 16 serum samples were detected to evaluate its significance in surveillance and follow-up after surgery. Results: The best model was composed by two protein peaks (3.9 KDa / BC1 and 5.6 KDa / BC2) with its sensitivity and specificity of 87.30% (55/63) and 95.00% (38/40) respectively. The sensitivity and specificity in the subsequent blind validation of new cases were 95.65% (22/23) and 85.00% (17/20) respectively. The diagnostic efficacies were the same to the patients of different stages (P>0.05). BC1's expression increased while BC2's decreased after operation (P<0.05). BC2's expression in the patients with recurrence or metastasis was higher than that in the tumor-free survivors (P<0.05). Conclusions: This method shows its potential in diagnosis, surveillance and follow-up after operation as well as screening of novel and better biomarkers in breast cancer. | |||

Abstract:Background and aims: The forkhead transcription factor Foxp3 is a key molecular which can mediate regulatory T cells immune-related inhibitory functions. Increased levels of FOXP3-positive Tregs in peripheral blood and focus of infection have been proved to be associated with a less favorable prognosis in various inflammatory diseases. It is of great interest to investigate the correlation between single nucleotide polymorphisms (SNPs) of FOXP3 gene and the susceptibility of Chronic Hepatitis B (CHB). Methods: Two SNPs rs2280883 and rs3761549 of FOXP3 gene in 344 patients with CHB and 372 matched controls were analyzed by Matrix-Assisted Laser Desorption/ Ionization Time of Flight Mass Spectrometry (MALDI-TOF). Results: At rs2280883 and rs3761549, there were no significant differences in the distribution of C and T alleles between CHB donors and healthy donors. Compared with healthy controls, patients with CHB had higher frequencies of TT genotype (74.1%) at rs2280883 or CC genotype (74.6%) at rs3761549 of FOXP3 gene. Patients who carried rs2280883 TT genotype (OR 1.1268 [95%CI 0.8097-1.5680], P<0.01) or rs3761549 CC genotype (OR 1.6291 [95%CI 1.1774-2.2541], P<0.00001) had high risk of suffering from CHB. Stratified analysis showed that rs2280883 TT/CT genotype was significantly associated with high incidence of HBeAg (P<0.01), and rs3761549 TT/CT genotype was also significantly associated with high incidence of HBeAg (P<0.01). Conclusions These results suggested that FOXP3 gene polymorphisms at rs2280883 and rs3761549 might be associated with the increased susceptibility to CHB. | |||

Abstract:The aim of this survey was to assess the prevalence and type distribution of oncogenic human papillomavirus (HPV) in women from Wufeng County within Hubei Province where the rate of cervical cancer mortality is the highest in China. Cervical DNA samples were collected from 1,100 women and then screened and quantified by multi-fluorescent, quantitative real time PCR assay. Furthermore, re-typed by real-time PCR using seven-pair primers of high- risk (HR) HPVs, designed to detect type 16, 31, 18/45, 33, 52, 58, and 67. The overall prevalence was 11.14% (CI 9.74-13.72%) in Wufeng County and no statistically different was found between Tu (12.17%, CI 10.03-12.34%) and Han nationality (8.95%, CI 3.55- 14.35%). Among different types of HPV, HPV16 was the most frequently-detected genotype and followed by HPV 52, HPV 58, HPV 18/45, and HPV 31. The most prevalent types in Tu nationality were HPV 16, HPV 52, HPV 58, but in Han nationality were HPV 16, HPV 33 and HPV 18/45. Meanwhile, the overall HPV prevalence was highest in the 20-24-year age group (21.43%, CI 17.56-25.30%) and also prevalent of HR-HPV was related with education. HPV 16,52, 58 are common genotypes in Wufeng County, as well as enhance the hypothesis that the second-generation HPV prophylactic vaccines including HPV 52 and 58 may offer higher protection for women in China and other Asian areas. Abstract:Notch signaling plays a critical role in determining cell fate such as proliferation, differentiation, and apoptosis. Accumulating evidences indicate that aberrant Notch signaling has tumor-promoting function in breast cancers. We hypothesized that Notch signaling may be a potential therapeutic target for human breast cancer. To address this issue, we down-regulated the expression of the Notch-1 receptor by siRNA in human breast cancer cells. We found that the down-regulation of Notch-1 signaling caused cancer cell growth inhibition by apoptosis induction. The effect of the down-regulation of Notch-1 may be through the inactivation of NF-κB. In addition, the down-regulation of Notch-1 signaling increased chemosensitivity to doxorubicin and docetaxel. Our results suggested that Notch signaling may be a promising target for breast cancer treatment. | |||

Abstract:Objectives: To investigate the clinicopathological features of anaplastic large-cell lymphoma (ALCL) and explore the optimal treatment strategies. Methods: Sixty-four primary systematic ALCL patients who were diagnosed and treated in our hospital were studied retrospectively. Results: Our data confirmed the male predominance, young age of onset and frequent presence of adverse factors such as B symptoms and involvement of extranodal sites in ALCL patients. However, patients with stage Ⅲ/Ⅳ disease and elevated LDH were less frequent in our study. Chemotherapy including CHOP and proMACE/cytaBOM was administrated in 46 and 9 patients, respectively. No significant difference can be seen in terms of response rate and survival between these two groups. Altogether, 16 patients received peripheral blood stem cell transplantation (PBSCT). Although significantly higher proportion of patients in transplantation group had state Ⅲ/Ⅳ disease, the long-term survival was comparable with that of patients received conventional chemotherapy. Anaplastic lymphoma kinase (ALK) was an independent prognostic factor, especially for adult ALCL patients, whose expression indicated significantly high long-term survival. Conclusions: CHOP is still the first-line chemotherapy regimen for ALCL patients due to its less toxicity and comparable efficacy with third-generation regimen. Patients with advanced disease may potentially benefit from PBSCT. | |||

Abstract:nvolvement of AFP against apoptosis of tumor cell has been implicated in its evasion of immune surveillance. However, the molecular events of immune escape mechanisms are still unknown. The major observations reported here relate to a possible mechanism by which heptoloma Bel 7402 cells escape immune surveillance in vitro. Western blotting and a well-characterized cofocal scanning image were performed to analyze the expression of Fas/FasL and caspase-3 in co-cultured Bel 7402 and Jurkat cells. After co-culture with Jurkat cells, up-regulated Fas and reduced FasL expression could be observed. Treatment with AFP could remarkably inhibit the elevated Fas and, whereas, induce the FasL expression in co-cultured Bel 7402 cells. Cells co-culture could induce the expression of caspase-3 in both cells line. The elevated caspase-3 in Bel 7402 cells was abolished following the treatment of AFP.
