Abstract:To investigate the relationship between SmCPS1 promoter polymorphism and the contents of tanshinones. We sequenced the SmCPS1 gene promoter region together with 388bp downstream from the translation start site and the content of tanshinones of 12 different germplasm of Salvia miltiorrhiza. To separate the functional polymorphic loci that may be related with the content of tanshinones from those not, the cis-elements of SmCPS1 promoter were predicted and analyzed by the Plant Transcriptional Regulatory Map database. We found 1) there was a different correlation between the polymorphism in the promoter region and the contents of the tanshinones. 2) Polymorphism on nine cis-elements, an 33bp insertion/deletion sequence and three tandem repeat variations in the sequence of SmCPS1 promoter may be related to the contents of tanshinones. These findings showed the relationships between SmCPS1 promoter polymorphism and tanshinones. | |||

Abstract:To investigate the mechanism of suppressive effect of Chinese medicine on Coronary Atherosclerotic Heart Disease (IHD) by observing the effect of Salvia miltiorrhiza injection coordinated with Acanthopanax senticosus injection(MSAI)on the acute myocardial ischemia rat. The optimal formula of MSAI was ascertained using design by uniform distribution. From pharmacodynamics, the rat was applied with isolated heart perfusion method, for measuring Coronary Artery Fluence . Further, Coronary Artery in rat was received physic liquor by intravenous injection using constant flow pump to determine left ventricular systolic pressure (LVSP) , left ventricular maximum pressure variation rate(LVMPVR)and left ventricular end-diastolic pressure(LVEDP). The MSAI was found that could significantly increase CF of the isolated heart of the rats(P<0.01),compared with SI and AI control group, respectively ; in addition,the effect of Formula (Ⅲ)(syringoside : protocatechuic aldehyde = 1:1.25)is the optimization. The results of hemodynamic effect of acute myocardial ischemia in 15min rat treated with MSAI demonstrated that MSAI could remarkably raise LVSP, LVMPVR(±dp/dtmax)and lower LVEDP. Additionally, the effect on resisting acute-myocardial-ischemia of MSAI group was better than that of SI group and AI group(P<0.01). The mechanism of suppressive effect of MSAI on IHD of may be realized by increasing CF, elevating LVSP, LVMPVR(±dp/dtmax),and lowering LVEDP. | |||

Abstract:The aim of the present study was to evaluate the diuretic effect on the urinary electrolyte concentration(Na+, K+, Cl-) and regulation of the relative mRNA expression of aquaporin-1(AQP1), aquaporin-2(AQP2), aquaporin-3(AQP3) and vasopressin V2 receptor (V2R)post oral administration of sclerotia of P.umbellata aqueous extract in normal rats. Aqueous extract of sclerotia of Polyporus umbellatus (50mg/kg, 250mg/kg, 500mg/kg) or the reference drug, furosemide (10mg/kg) were administrated orally to male SD rats and their urine output was quantified and collected 24h and 8days after the treatment. The kidney medulla AQP1, AQP2, AQP3 and V2R mRNA relative expressions were measured with RT-PCR. After single dose of the exact of sclerotia of P.umbellata, urine output was found to be significantly increased, which began at 4h, and at 24h after the treatment, the sclerotia of Polyporus umbellatus extract and furosemide treatment produced the similar total volume of urine excreted. The extract increase urinary levels of Na+, K+, Cl-, to about the same extent, while furosemide increased urinary levels of Na+ and Cl-. After the 8-day doses, all two substances induced significant diuresis, natriuresis and chloriuresis. These two substances don’t regulate the AQP1 and AQP3 mRNA level in normal rat kidney medulla. The AQP2 mRNA level of sclerotia of P.umbellata extract was down-regulated significantly, the V2R mRNA level of sclerotia of P.umbellata extract 50mg/kg dose group and 250mg/kg dose group were down-regulated significantly too. Interestingly, the low-dose group had higher effect on regulation of AQP2 and V2R mRNA level. Aqueous extract of sclerotia of Polyporus umbellatus has conspicuous diuretic effect confirming its ethnopharmacological use. From the pattern of excretion of water, sodium, potassium, cholorine, AQP2 and V2R mRNA level, it may be logically concluded that it has effect from down-regulating AQP2, and down-regulate AQP2 by down-regulating V2R. | |||
