Abstract:Long non-coding RNAs (lncRNAs) have been reported to be involved in processes of tumor progression. 1α,25-dihydroxyvitamin D3 (1α,25(OH)2D3), the active form of vitamin D, has an anti-proliferative effect on tumor cells. Therefore, differentially expressed lncRNAs regulated by 1α,25(OH)2D3 may be associated with tumor progress. This study was aimed to investigate the lncRNAs profiling pattern regulated by 1α,25(OH)2D3 in ovarian cancer cell line SKOV-3 using microarray analysis. After 72 hours of 1α,25(OH)2D3 treatment, 139 lncRNAs and 105 mRNAs were significantly dysregulated. Five lncRNAs were validated by quantitative reverse transcription polymerase chain reaction (qRT-PCR). In functional analysis, dysregulated lncRNAs can participate in biological processes related to systemic lupus erythematosus, alcoholism, osteoclast differentiation and cancer signaling pathways, such as Wnt, TNF and TGF-β receptor signaling. The dysregulated lncRNAs were associated with their co-expressed genes and may be mainly regulated by these three transcription factors, TAF7, POU2F2 and KAT2A. Additionally, there were 17 lncRNAs co-expressed with genes VDR, CYP24A1 and CDH1, suggesting that these lncRNAs acted as the connection to associate vitamin D signaling with EMT. These data constitute the first report of lncRNA expression patterns regulated by 1α,25(OH)2D3 in human ovarian cancer cells, and will provide new evidence for the anti-tumor effect of 1α,25(OH)2D3 through lncRNAs. | |||

Abstract:Increasing evidence has shown that epithelial-mesenchymal transition (EMT) can endow cancer cells with motile and invasive properties. In ovarian tissue, EMT not only plays a physiological role in the postovulatory repair of ovary surface epithelial (OSE) cells, but also an important role in the aggressiveness and recurrence of ovarian cancer. Previous research has indicated that 1α,25(OH)2D3 decreases the migration and invasion of many kinds of tumor cells by suppressing EMT. However, it remains unclear whether 1α,25(OH)2D3 can inhibit the process of EMT in mouse OSE (MOSE) cells at different stages of oncogenic transformation. In this study, we found that 1α,25(OH)2D3 significantly reduced the migration and invasion of late malignant transformed MOSE (M-L cells) cells by inhibiting EMT both in vitro and in vivo, but not in intermediate transformed (M-I) cells. Interestingly, the expression of CYP24A1 in M-I cells was distinctly higher than M-L cells following continuous treatment with 1α,25(OH)2D3. Furthermore, we demonstrated that the knockdown of CYP24A1 enhanced the anti-invasion properties of 1α,25(OH)2D3 on both M-I and M-L cells by upregulating the expression of E-cadherin, and downregulating the expression of N-cadherin, Vimentin, β-catenin and Snail. These findings imply that CYP24A1 is a potential target for the inhibition of EMT and ovarian cancer metastasis. | |||

Abstract:1,25(OH)2D3 deficiency may be associated with affective disorders, Parkinson's disease, Alzheimer's disease and cognitive abilities. To explore the protective effects of 1,25(OH)2D3 against glutamate-induced damage in a mouse-derived hippocampal HT-22 cells and underlying potential mechanism. The cell survival rate was determined with Cell Counting Kit-8. Cell cycle, apoptosis and reactive oxidative species (ROS) release were detected by FACs technology. Results showed that pretreatment 1,25(OH)2D3 effectively increase HT-22 cell viability up on glutamate exposure. This might be associated with the fact that 1,25(OH)2D3 significantly reduced the percentage of G0/G1 circles (G0/G1%) (p<0.01),increased the proportion of S phase (S%) and G2/M phase (G2/M%) cells (p<0.01). Moreover, apoptosis in HT-22 cells showed the late cell apoptosis rate was decreased (p<0.05) in glutamate group incubated with 1,25(OH)2D3 (i.e. combination group) compared with the glutamate only group. There was an inverse association between the concentration of 1,25(OH)2D3 and apoptosis rate. Meanwhile, the ROS release was elevated in the combination group (p<0.05). Our study indicates that 1,25(OH)2D3 might adversely affect glutamate-induced apoptosis in the HT-22 cells,this might be owing to the fact that 1,25(OH)2D3 could increase cell proliferation and decrease late apoptosis rate. 1,25(OH)2D3 might be a promising therapeutic agent for the treatment of depression. | |||

Abstract:Green tea possibly elicits protective effects against colorectal cancer. Thus, we conducted a meta-analysis of prospective cohort studies to evaluate whether green tea has a preventive effect against colorectal cancer. A systematic literature search was conducted in PubMed. Prospective cohort studies that reported risk ratios (RRs) with 95% confidence intervals (CIs) of colorectal cancer risk of green tea consumption were included. Either a fixed-effects or a random-effects model was used to calculate pooled effects. Combined relative risk (RR) ratios for the highest vs. lowest were calculated. The combined RR derived from 10 cohort studies was 0.89 (95% CI: 0.78-1.00; p<0.001), which show that green tea consumption decreased the risk of colorectal cancer. Subgroup analysis suggested that green tea consumption significantly decreased the risk of colorectal cancer in women(RR, 0.85; 95% CI, 0.72-0.99). In summary, green tea consumption has favorable effects on colorectal cancer, especially in women. However, the result should be interpreted with caution because of the evidence of heterogeneity. | |||

Abstract:In this paper, we conducted a case control-based genetic screening of 14 single nucleotide polymorphisms (SNPs) loci in five genes including thyroglobulin (TG),thyroid peroxidase (TPO), thyroid stimulating hormone receptor (TSHR), dual oxidase 2 (DUOX2) and solute carrier family member 5 (SLC5A5), with the aim to explore the relationship between the candidate SNPs and the susceptibility to hypothyroidism(HT) in Chinese Han population. A total of 347 hypothyroid patients and 337 healthy individuals were enrolled and the SNP loci were sequenced using Sequenom platform and MassArray technology.Allele association analysis showed rs2741202 and rs3739266 in TG were associated with HT (P = 0.048 and 0.038, respectively). Genotype association analysis showed rs1126799 in TPO and rs2741202 in TG had weak association with HT (P = 0.067 and 0.098, respectively). Logistic regression analysis showed the females were more susceptible to HT than the males (OR = 0.247), and individuals carrying the mutation genotype of TG-rs3739266 (OR = 1.372) were more susceptible to HT than those with the wild genotype. SLC5A5-rs11672428 was associated with FT4 after the adjustment of age and gender (P = 0.003), and TG-rs3739266 was associated with TSH in the females (P = 0.028). These SNP loci may be valuable biomarkers for the screening of the susceptible population. | |||

Abstract:To investigate the interaction effects of promoter region A163G,T245G and T950C polymorphisms of osteoprotegerin(OPG) gene and calcium intake on bone mass in premenarche girls. Methods: The OPG genotypes were determined by PCR-RFLP in 214 premenarche girls of Han race in Guangzhou area, and bone mineral density(BMD) at lumbar spine(L1-4), femoral neck, Ward’s triangle area(WSBMD) and total BMD, were measured by dual-energy X-rayabsorptiometry(DEXA). Current dietary calcium intakes were assessed by 3-day food record questionnaire and 3-day weighed food record method. Results: Hardy-Weinberg equilibrium was evident for OPG polymorphisms. After adjusting the confounding factors of year, height and weight, ANCOVA showed significant effect of interaction of calcium intake and T950C SNPs on WSBMD(P=0.046). WSBMD of subjects in calcium intake<500mg/d group was lower 13.0% 、23.7% than those in 500~800mg/d group and >800 mg/d group respectively among girls with TT genotype of T950C locus. However, such significance was not detected among girls with TC or CC genotype of T950C locus. Significant interaction was not observed between A163G、T245G SNPs and calcium intake on BMD. Conclusion: calcium intake and OPG T950C SNP has significant interaction on WSBMD in premenarche girls. More calcium intake would be benefit to gain higher WSBMD for girls with TT genotype of T950C locus than those with TC or CC genotype | |||
