Abstract:Chinese five element theory (CFET) has been a long-standing puzzle in both Chinese Medicine and Western Medicine. The scientific explanation of the theoretical basis of CFET has been a difficult challenge for scientists. In particular, the composition, mutual promotion and mutual restraint of the five elements of CFET have been the focus of scientific debates. How to use scientific methods to clarify the philosophical principles is the urgent problem to be solved. CFET was discovered by the ancient observing the celestial naked-eye planets and their movement rules. The present study aims to explore the relationship between CFET and the operation of solar planets using orbital mechanics and the information entropy theory. Here, the position vector and velocity vector of the solar planets Mercury, Venus, Earth, Mars, Jupiter and Saturn were calculated based on orbital mechanics, and then the angles in the triangles composed of any three planets were calculated and analyzed. The results of information entropy analysis showed that Jupiter, Mars, Venus and Mercury could build the prototype of the five element theory, and the introduction of Earth could further improve and perfect the theory of the five elements. This papre presents that the motion law of Jupiter, Mars, Venus and Mercury, which could lay the foundations for CFET, also naturally explains CFET's development context and meaning. CFET is closely related to the five solar planets Jupiter, Mars, Earth, Venus and Mercury, could construct a self-organizing system with mirror symmetry breaking, and may reveal the nature of the origin of all things.. | |||

Abstract:Objective: To explore the rationality of herbaceous compatibility of peony liquorice decoction by peoniflorin and glycyrrhetinic acid's pharmacokinetic parameters in SD rats' plasma. Methods: A HPLC/MS/MS method was used to detect the concentration of peoniflorin and glycyrrhetinic acid in SD rats which were administrated with peony, liquorice or peony liquorice decoction separately. Results: Comparing with the single administration of peony or liquorice, the rats given with peony liquorice decoction the glycyrrhetinic acid in the plasma spent less time to reach the peak concentration and the Cmax also increased. What's more, the peoniflorin's peak concentration enhanced, and the relative bioavailability increased either. Conclusion: The liquorice promoted the absorption of peoniflorin in peony, enhanced its vitro concentration, thereby causing the peoniflorin's function stronger and more powerful. The peony has advance the time of glycyrrhetinic acid in liquorice and quantity which appears in intracorporal, causing its effect emerged rapidly. | |||

Abstract:[Object]To unveil potential biologic mechanisms between Qi and blood through analysis of Si-Wu-Tang (SWT) and Si-Jun-Zi-Tang (SJZT) with biomedical approaches. [Methods]Ingredients of eight herbs from SWT and SJZT were retrieved from TCMID database. The genes targeted by these ingredients were chosen according to their STITCH defined high confident (combined-score more than 0.7). We then performed enrichment analysis by Gene Ontology (GO) and DAVID. Enriched GO terms with p-value less than 0.01 were collected and interpreted the biological meanings through DAVID and KEGG. Then, a visualized network was constructed with ClueGO (a Cytoscape plug-in) to compare the difference pathways between SWT and SJZT formulae. [Results] A total of 243 genes targeted by 195 ingredients of SWT formula and 209 genes targeted by 61 ingredients of SJZT were obtained by retrieving from TCMID database. Six metabolism pathways and two environmental information processing pathways enriched by targets were correlated with two or more herbs in SWT and SJZT formula, respectively. Expect for disease associated pathways, SWT significantly influence amino acid and carbohydrate metabolism. While pathways enriched by genes targeted by ingredients of SJZT mainly belonged to organismal system function, environmental information processing, metabolism and cellular processes. The common pathways of SWT and SJZT belonged to organismal systems, metabolism, environmental information processing and cellular processes. [Conclusion]SWT with the functions of influencing amino acid and carbohydrate metabolism is significant different from SJZT with the actions of influencing neuroendocrine system by affecting serotonergic synapse to regulate thyroid hormone, ovarian steroidogenesis, prolactin and oxytocin secrction and the actions of regulating corrective hormones secretion and promoting vascular smooth muscle contraction. The common effects of SWT and SJZT are regulating the functional activities of the hypothalamic pituitary adrenal and gonadal axis by affect inhibitory synapses (GABAergic synapse) to stimulate estrogen and steroid hormones secretion and strengthening the ability of anti-infection. All the different and common pathways also reflect the characteristics of blood deficiency and Qi deficiency and indirectly reflect the molecular mechanism of blood and Qi of TCM.) | |||

Abstract:Objectives: Zi-Yin decoction is a herbal formula in traditional Chinese medicine used for the clinical treatment of polycystic ovary syndrome (PCOS). This study was aimed to investigate whether Zi-Yin decoction had an impact on follicular development of PCOS through stem cell factor (SCF). Materials and methods: The PCOS model was induced by dehydroepiandrosterone (DHEA). Granulosa cells (GCs) were collected and co-cultured with 10%Zi-Yin medicated serum (ZMS), 20%ZMS or recombinant follicle stimulating hormone (FSH) for 24, 48 and 72 hours in vitro. The concentration of SCF and estradiol (E2) in culture fluid was measured by ELISA. The expression of SCF gene was analyzed at mRNA by real-time RT-PCR. Results: DHEA-induced rats with PCOS exhibited significantly lower level of SCF in GCs at mRNA level after cultured in vitro for 72 hours (P<0.05 versus controls). Treatment with either 10% or 20% ZMS attenuated the secreting dysfunction induced by DHEA-exposed, as evidenced by a increasing of SCF, plus E2 or not, in GCs from antral follicles (P<0.05 versus DHEA). Likewise, culture with FSH promoted the expression of SCF mRNA and concentration of E2 and SCF in culture fluid. This effect of FSH could be enhanced by addition with 10%ZMS. Conclusions: Decreased in SCF expression may occur in patients with PCOS and result in abnormal sex hormone secretion of GCs. The herbal formula treatment of Zi-Yin decoction will be beneficial for patients with PCOS via inducing SCF expression of antral follicles to promote follicular maturation.????? | |||

Abstract:Although acupuncture has been partially recognized for its therapeutic functions in Western countries, the meridian theory is still highly disputed. So far, no substantial anatomical structure has been found to support the existence of "Jing" or "channels" and "Luo" or "collaterals". The regulatory function of the meridians cannot yet be explained based on modern biology. In this article, the meridian system was compared with nematode nervous system, and it was found that there were high structural similarities between the two systems. Therefore, based on the biogenetic law that animal ontogeny is simply a quick recapitulation of phylogeny, I hypothesized that the meridian was originated from the nematode nervous system which was recapitulated during the human embryonic development, and meridian cells were the residual cells of nematode nervous systems. Here it was shown that many meridian phenomena can be well explained by the hypothesis, such as the bidirectional signal transduction, surface vs. viscus correspondence, delay of effect, linear features, stimulus strength threshold and diversity of stimuli. In addition, it can also explain other meridian phenomena, such as "De Qi " or "engaged", sensation along the meridians, meridian-specific skin disease, low resistance and high calcium ion concentration at the acupoints, high temperature and lighting along the meridians. I also proposed a number of ways to validate the hypothesis for two key aspects: (1) the truthfulness of the meridians, (2) the origin of meridian cells. For example, induced pluripotency technology can be used to demonstrate the embryogenicity of the meridian cells, and photogenetics techniques can be used to verify the nervous properties of the meridian cells.. | |||

Abstract:Objective: To investigate the molecular mechanisms behind the therapeutic effects of Duhuo Jisheng Decoction (DHJSD) on osteoarthritis (OA). Methods: A total of 27 two-month-old male Sprague Dawley rats were randomly divided into three groups: the control group (no papain-induced OA; received an equivalent amount of saline only), the model group (papain-induced OA; received an equivalent amount of saline only) and the DHJSD group [papain-induced OA; received a clinical oral dose of DHJSD (9.3g/kg/day)]. After 8 consecutive weeks of treatment, the morphological changes in articular cartilage were observed under an optical microscopy and by transmission electron microscopy (TEM), and the mRNA and protein expression levels of Cyclin D1, CDK4, CDK6, Rb and p16 were measured by RT-PCR and immunohistochemistry, respectively. Results: Treatment with DHJSD significantly improved the arrangement of articular cartilage structure and collagen fibers and reduced cell degeneration compared with the model group. The mRNA and protein expression levels of Cyclin D1, CDK4, CDK6 and Rb in DHJSD-treated group were significantly increased compared to those in the model group, whereas p16 expression was significantly down-regulated. Conclusion: DHJSD treatment promotes chondrocyte proliferation by promoting the G1/S checkpoint transition in the cell cycle and by up-regulating the expression of Cyclin D1, CDK4, CDK6 and Rb and down-regulating the expression of p16 and this may, in part, explain its clinical efficacy in the treatment of osteoarthritis. | |||

Abstract:Background:This study investigated the dual control mechanism of kidney nourishing therapy modulating the cell cycle in Lewis-bearing mice with cyclophosphamide induced myelosuppression. Methods: Thirty Lewis-bearing mice were randomly grouped into an untreated group, control group, and treated group. Both treated and untreated groups were intraperitoneally injected with cyclophosphamide to produce a myelosuppression model. Mice in the treated group were fed with theShuanghuangShengbaigranule (40 g/day) for 6 consecutive days. Standard blood tests and the count of bone marrow nuclear cells were performed, and the cell reproductive cycles of bone marrow and tumorswere measured in these mice. In addition, the western blot approach was used to measure the upstream activating signals of CyclinD-CDK4/6 such as c-Myc and CDC25A, the upstream suppression signals such as p16INK4a and p15INK4b, and the expression of downstream activated signals such as Rb, pRB, and E2F. All of the tested results were validated by reverse transcription quantitative real-time polymerase chain reaction. Results: The results showed that the ShuanghuangShengbaigranule could elevate the count of leukocyte and bone marrow nuclear cells of Lewis-bearing mice with cyclophosphamide induced myelosuppression. It could also stimulate bone marrow cells to move from G0/G1 phases to S phase, accelerating the progress of the cell reproductive cycle and increasing the cell proliferation index. Simultaneously, the ShuanghuangShengbaigranule could also suppress cancer cells moving from G0/G1 phase to S phase, reducing the proliferation index. The tumor weight of Lewis-bearing mice in the treated group was much less than those of the control group. Expression levels of c-Myc, CDC25A, Rb, pRb, and E2F of bone marrow in ShuanghuangShengbaigranule-treated mice was higher compared to the control group, whereas they were lower in the cancer cells. Conclusion: The experimental results demonstrate that the ShuanghuangShengbaigranule has dual control on the cell reproductive cycles in cancer cells and bone marrow nuclear cells in Lewis-bearing mice. | |||

Abstract:Purpose This study aimed to evaluate the efficacy and safety of traditional Chinese surgical treatment "suture dragging combined with pad compression" vs fistulotomy for anal fistulae with secondary tracks and abscess. Methods From January 2008 to March 2009, 60 patients with intersphincteric or transsphincteric anal fistulas with secondary tracks and abscess were randomly divided into study group (suture dragging combined with pad compression, SDPC) and control group (fistulotomy, F). The time of healing, recurrence rate, postoperative pain (VAS), patient satisfaction,incontinence evaluation and anorectal manometry before and after treatment were all examined. Results The time of healing (SDPC 24.33d vs F 31.57d; p<0.01) was significantly shorter and patient satisfaction score (SDPC 4.07 vs F 3.37; p<0.05) was significantly higher in the SDPC group. There were no significant differences between both groups regarding age, gender, fistulae type, mean postoperative pain scores, recurrence rate, incontinence evaluation and anorectal manometry (including maximal anal squeeze pressure and anal resting pressure) before and after treatment. Conclusions Traditional Chinese surgical treatment "suture dragging combined with pad compression" for anal fistulae with secondary tracks and abscess is less invasive, safe and effective. | |||

Abstract:Aim: This study was designed to develop and treat with traditional Chinese medicine method of an experimental rat model of subcutaneous fistula induced by bacteria infection. Methods: A spring-gauze was surgically implanted into 24 male Sprague-Dawley (SD) rat's dorsal muscle, respectively. Then Staphylococcus aureus and Escherichia Coli mixed bacteria were injected into the spring-gauze. After 45days, all spring-gauzes were removed under anesthesia. X-ray and ultrasound were performed, enabling imaging fistula track assessment. Every rat was examined by a fistula probe and four rats were randomly selected for histological fistula tract assessment at the same time. Residual rats were randomly divided into in two groups. Group A treated with traditional Chinese medical method "thread-dragging therapy". Group B treated with Western medical method "fistulotomy". The healing time and body weights of two groups were compared. Results: At 45days imaging assessment showed a track with two openings in all rats and histological results proven lumen and surrounding granulation tissue with epithelium similar to that criteria and features of fistula in human being. The mean healing time of group A was shorter than group B with significant differences. The body weight of group A is heavier than group B at 7days after surgery and 14days after surgery respectively. Conclusions: A rat model with subcutaneous fistula induced by Staphylococcus aureus and Escherichia Coli mixed bacteria infection is easy and repeatable.
