Scutellariae Radix is a Chinese herb for detoxification. Efficient extraction of its main active components baicalin and saccharides is interesting to pharmaceutical and cosmetic fields. The baicalin and saccharides were efficiently extracted with aqueous two-phase systems (ATPSs) Ethanol-(NH4)2SO4-H2O (EtSO) and Ethanol-cosolvent-(NH4)2SO4-H2O (EtSO-cosolvent). The factors such as pH, (NH4)2SO4 concentration, ethanol fraction, cosolvent fraction, extraction time and temperature were optimized and thermodynamics for partition was discussed. With the extractions at 48 C for 40 min, the EtSO, EtSO-MtOH and EtSO-Acetone exhibited higher baicalin yield than conventional 70% ethanol system (v/v) by 21.4%, 24.8% and 26.2%, respectively; and they displayed saccharides yield in a similar level to that with water system. By dialysis method the saccharides extracted with ATPS was proved to have similar molecular weight to those extracted with water. Therefore, it is feasible to obtain two birds (two targets) with one stone (ATPS) in high efficiency.
