High molecular weight (HMW) glutenin subunit compositions and gliadin variations in 91 cultivated emmer wheat (Triticum turgidum ssp. dicoccum) were analyzed by sodium-dodecyl-sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and acid polyacrylamide gel electrophoresis (A-PAGE), respectively. In the HMW glutenin subunits, four and nine allelic variants were detected at the Glu-A1 and Glu-B1 loci, respectively. Alleles Glu-A1c (47.3%) and Glu-B1b (35.2%) were the most frequent at the Glu-A1 and Glu-B1 locus, respectively. At Glu-1 loci, a total of 24 combinations were obtained, among which Glu-A1c/Glu-B1b was the main combination. A total of 58 gliadin bands were detected in all accessions, resulting in 84 gliadin patterns. All materials were clustered into three groups, and indicated that the genetic diversity in dicoccum wheat was associated with its geographical distribution. |
