DNA topoisomeraseⅡα(TOPOⅡα) is a nuclear enzyme involved in chromosome segregation and other essential cellular processes. IHC and Western blot were used to determined the changes of TOPO II a expression and of its behavior in HeLa cells irradiated withγ-rays(≤2 Gy). A transient and significant decrease of TOPOⅡαafter low doseγ-rays exposure was detected: the decrease began at 4h after irradiation, reached to the lowest level at 12h, and began to return at 16h. Given low dose γ-rays, the level of decrease added as dose was added. The level of decrease after multiple irradiation was higher than that after single irradiation. This observation indicates that the decrease of TOPOⅡαafterγ-rays is time-dependent and dose-dependent, and the effect of γ-rays can add up. So the change of TOPOⅡαexpression may be related to chromosome non-disjunction.
