This paper described a practical five-step synthetic approach for 2-amino-5-methoxylpropiophenone in 45% overall yield. Thus, the starting material 3-chloropropiophenone was first subjected to nitration to give compound 9, which was protected with glycol to give compound 10. Compound 10 was subjected to nucleophilic aromatic substitution, deprotection and reduction to afford the target compound. The advantages of this procedure include short reaction steps, simple operation and good yield.
