To investigate the effect of glutamine(Gln) on the expression of grp75 and the protective effect in PC12 cell line under glucose deprivation (GD), grp75 expression of PC12 cell with administration of Gln was analyzed by western blot and immunocytochemistry, and the change of mRNA level of grp75 was detected by RT-PCR. The cell viability (survival and apoptosis rate) and mitochondria function (mitochondria transmembrane potential) were also observed by MTT method and flow cytometry. The results showed that Gln could enhance the expression of grp75 at both a protein level and a mRNA level. The cell treated with Gln, its survival rate increased and apoptosis rate decreased under glucose deprivation. These data suggested that glutamine can protect cells from the injury induced by glucose deprivation through the expression of grp75.
