Objectives: Zi-Yin decoction is a herbal formula in traditional Chinese medicine used for the clinical treatment of polycystic ovary syndrome (PCOS). This study was aimed to investigate whether Zi-Yin decoction had an impact on follicular development of PCOS through stem cell factor (SCF). Materials and methods: The PCOS model was induced by dehydroepiandrosterone (DHEA). Granulosa cells (GCs) were collected and co-cultured with 10%Zi-Yin medicated serum (ZMS), 20%ZMS or recombinant follicle stimulating hormone (FSH) for 24, 48 and 72 hours in vitro. The concentration of SCF and estradiol (E2) in culture fluid was measured by ELISA. The expression of SCF gene was analyzed at mRNA by real-time RT-PCR. Results: DHEA-induced rats with PCOS exhibited significantly lower level of SCF in GCs at mRNA level after cultured in vitro for 72 hours (P<0.05 versus controls). Treatment with either 10% or 20% ZMS attenuated the secreting dysfunction induced by DHEA-exposed, as evidenced by a increasing of SCF, plus E2 or not, in GCs from antral follicles (P<0.05 versus DHEA). Likewise, culture with FSH promoted the expression of SCF mRNA and concentration of E2 and SCF in culture fluid. This effect of FSH could be enhanced by addition with 10%ZMS. Conclusions: Decreased in SCF expression may occur in patients with PCOS and result in abnormal sex hormone secretion of GCs. The herbal formula treatment of Zi-Yin decoction will be beneficial for patients with PCOS via inducing SCF expression of antral follicles to promote follicular maturation.?????
