he present studies described characterization of two rice bentazon-lethal mutants, bel1 and bel2, which were created via γ-ray radiation in an indica photo-thermo-sensitive genic male sterile (P/TGMS) line, W6154S, and a japonica inbred line, Norin8, respectively. Both mutants of bel1 and bel2 started to show affected symptoms at a concentration of 100 mg/L and became lethal at 300 mg/L bentazon but within this range bel1 is slightly more tolerance than bel2. The lowest concentrations of their bentazon-sensitivity, however, were about 120-fold lower than those of their wild controls. Using bel1 to cross with bel2 followed by herbicide test of their progenies, we confirmed that these two mutants were genetically allele each other. The data from PCR amplification followed by sequence comparisons between wild and mutant alleles revealed a single base-pair deletion in each mutant and for bel1 this occurred at 408th bp in its second exon, while for bel2 this at 507th bp in its first exon. |
