Abstract:Deer antlers are the only mammalian appendages to display annual cycle of full regeneration. However, little is known about the molecular mechanisms of antler regeneration. Our previous study has demonstrated that parathyroid hormone-related peptide (PTHrP) can promote proliferation of antler chondrocytes and inhibit its differentiation. But the mechanism underlying such regulation is not fully understood. The present study aimed to determine the role of PTHrP on the mRNA expression of matrix metalloproteinase-13 (MMP13) in the antler chondrocytes. The possible pathways that transduce PTHrP effects were also investigated in the cells. The in situ hybridization results showed that MMP13 were mainly localized in the dermal fibroblasts, perichondrium and cartilage in the sika deer antler, of which MMP13 were highly expressed in the antler chondrocytes. Exogenous PTHrP could inhibit the expressions of MMP13 in the antler chondrocytes. The inhibitory effect of PTHrP on MMP13 was eliminated by P38MAPK inhibitor SB203850 and PKC inhibitor GF109203X. These results suggest that PTHrP can inhibit MMP13 expression by p38MAPK and PKC signaling pathways in the antler chondrocytes, thus PTHrP is involved in the control of antler chondrocytes maturation and cartilage matrix degradation. | |||

Abstract:The present study was to identified the expression of Ghrelin and GHS-R in mouse thymic medullary-type epithelial cells (MTEC1).. Whether the gene of Ghrelin and GHS-R are expressing in MTEC1 cells or not was measured by using quantitative real time RT-PCR (Q-PCR). Results identified that the mRNA of Ghrelin and GHS-R can be detected in MTEC1 cells. The protein levels of GHS-R and ghrelin precursor were detected by the method of western blot analysis. Moreover, the location of GHS-R in MTEC1 cells were detected by immunofluorescence and the quantity of ghrelin in the supernatant of MTEC1 cells were measured by ELISA. The results of western blot analysis make sure that GHS-R and precursor of ghrelin are expressing in MTEC1 cells. GHS-R is a kind of transmembranes protein. as the results of immunofluorescence shown The quantity of ghrelin in the supernatant of MTEC1 cells measured by ELISA was 39.20 pg/ml. | |||

Abstract:Toll-like receptor 4 (TLR4) has been suggested to play a very important role in modulation of immunological tolerance in the human female reproductive tract. To better understanding of functions of TLR4 in establishment and maintenance of pregnancy in pigs, the study was performed to compare the expression patterns of TLR4 at maternal-fetal interface of pregnant Yorkshire and Meishan pigs. The uteri from Yorkshire and Meishan gilts on days 26 and 50 of gestation were obtained respectively. Immunohistochemical analysis indicated that TLR4 was detected in uterine epithelium and trophoblast of these two types of pigs on days 26 and 50 of gestation. In Yorkshire gilts, the expression of TLR4 was higher in uterine epithelium than that in trophoblast on day 26 of gestation, while it was lower in uterine epithelium than that in trophoblast on day 50 of gestation. In Meishan pigs, the expression of TLR4 in trophoblast was higher than that in uterine epithelium on either day 26 or day 50 of gestation and TLR4 was found to be mainly present in the cells located at the bottom of trophoblast on day 50 of gestation. Compared with Meishan pigs, TLR4 was abundant in uterine epithelium of Yorkshire pigs on days 26 and 50 of gestation, yet it was decreased in trophoblast of Yorkshire pigs on day 50 of gestation. The results indicated that TLR4 at the maternal-fetal interface is expressed in differential patterns between Yorkshire and Meishan pigs and provided novel information about the functions of TLR4 in pig pregnancy. | |||
