Abstract:Collecting Covid-19 genomes from three regions: Shanghai-China, Tbilisi-Georgia and Sydney-Australia, five similar genomes are selected from each region for research in this paper. Applying ”Datum gene sequence” method proposed,our results are shown that variation is immense in Sydney-Australia region, then variation is shown in the second in Tbilisi-Georgia region and it has a minimal value in Shanghai-China region respectively. | |||

Abstract:Recently, urine-derived stem cells (USCs) have shown application potential for wound healing based on cell therapy strategy. However, the mechanism remains unclear. As it has been reported the interactions between stem cells and recipient cells in wound sites can stimulate wound healing through paracrine effects, we hypothesized that the wound healing ability of USCs might be related to paracrine effects between USCs and recipient cells involved in wound healing, mainly including endothelial cells and fibroblasts. In addition, our previous studies demonstrated that bioactive silicate materials could stimulate paracrine effects between bone marrow stem cells and endothelial cells as well as fibroblast and endothelial cells, which finally enhanced vascularization in bone regeneration. Therefore, in this study, we aimed to investigate the paracrine effects between USCs, human umbilical vein endothelial cells (HUVECs) and human dermal fibroblasts (HDFs) and to study the effects of bioglass (BG) on these paracrine effects in order to explore the application potential of BG combining USCs on wound healing. Results show that BG ion extracts can not only affect behaviors of USCs but also stimulate paracrine effects between USCs and HUVECs, USCs/HDFs and USCs/HUVEC-HDF co-cultures, which results in stimulated vascularization of HUVECs, promoted ECM protein production and myofibroblast differentiation of fibroblasts either in indirect contact co-cultures or in USC-HUVEC direct contact co-cultures. Taken together, USCs possess application potential in wound healing based on cell therapy due to its strong effects on fibroblast and endothelial cells. More importantly, BG can be used to promote wound healing ability of USCs in tissue engineering therapy though enhancing the interactions between USCs and other types of cells. | |||

Abstract:Microbial populations in fermented grains during fermentation is the key to influence the flavor of Chinese liquor. In this paper, fermented grains samples from Hetao were used for DNA extraction, we studied the effects of six different extraction methods on total DNA extraction from fermented grains. The follow-up experiment showed that the obtained results in the use of the repeated freezing and thawing was the best for DNA extraction and polyacrylamide gel electrophoresis. Comparing with the other methods, the repeated freezing and thawing can reflect better the diversity of microorganism. | |||

Abstract:CVB3 capsid protein-1 (VP1) is an immunodominant structural protein that plays an important role in eliciting of host immune response. However, the termini of linear proteins are often flexible and easily become the targets of proteolytic enzymes. An alternative way to overcome the limitations of linear protein is the cyclization of the polypeptide backbone. It has been shown that circular proteins can increase their thermostability and resistance to chemical, thermal, or enzymatic degradation due to lacking of both amino and carboxyl termini. In this study, we have joined the N- and C-termini of VP1 protein with a normal peptide bond using intein-mediated protein trans-splicing, which produced the VP1 protein with a circular topology. Successful cyclization was proven by relinearization by proteolytic cleavage and by resistance against carboxypeptidase. The circular form of VP1 was found to be significantly more stable against irreversible aggregation upon heating than the linear form. | |||

Abstract:As the important structure of oocyte and early embryo, zona pellucid causes adverse effect to the development of the embryo because of thickness and hardening during development. The present study examined the effect of zona pellucid for porcine embryo development and the influence of electric stimulation and culture systems (WOWs type) for ZP-free embryo development. ZP-free oocytes were parthenogenetically activated (PA) with a single direct current (DC) pulse of 0.86 kV/cm for 80 μs, while ZP-intact oocytes activated with a single DC of 1.26 kV/cm or 0.86 kV/cm for 80 μs, followed by 4 h treatment with porcine zygote medium 3 (PZM-3) added 5 μg/ml cytochalasin B and 10 μg/ml cycloheximide. The results showed that the developmental competence of ZP-free and ZP-intact PA embryos was corresponding when proper electrical strengths with a DC for 80 μs of 0.86 kV/cm and 1.26 kV/cm were executed respectively. To investigate the effect of WOWs' shape and size on the developmental viability of ZP-free PA embryos, the matured ZP-free PA oocytes were randomly grouped and cultured into three type WOWs: S-W: the small bowl-shaped WOW; L-W: the large bowl-shaped WOW; VF-W: the V-shaped but flat bottom WOW. The rate of 2-cell was no different in the three types WOWs (p > 0.05), but more embryos cultured in S-W (76.4±4.4%) will developed into 4-cell stage compared to embryos cultured either in L-W or in VF-W (61.6±2.1% or 63.0±4.7%). The blastocyst rate showed a significantly decreasing tendency from 50.2±6.4% (S-W) to 31.2±3.4 %(L-W) and 13.0±5.1 %(VF-W). To assess the effect of ZP on the hatching ability and the survival viability, blastocysts from ZP-free and ZP-intact pig PA embryos were collected on Day 7 and were transferred into a new Nunc 4-well dish in PZM-3. The survival viability of ZP-free blastocyst cultured to day 8, 9 and 10 was comparable to ZP-intact blastocyst. But the ZP-intact blastocyst hatched rate was very low with only 17.9±5.7%. Accordingly, the presence of zona pellucida is not essential under certain in-vitro culture situations using the WOW culture system and may harmer the hatching viability of ZP-intact porcine blastocysts cultured in vitro. | |||

Abstract:Penicillium purpurogenum Li-3, a fungus producing β-glucuronidase (PGUS), can modify glycyrrhizin (GL) to glycyrrhetinic acid monoglucuronide (GAMG) when grown in medium with GL as the sole carbon source. In order to improve the conversion rate of GL to GAMG and the yield of GAMG, licorice extract (LE) was added as an inducer to enhance the production of PGUS, and the influence of LE on the conversion rate of GL to GAMG was studied in this work. When the strain were grown in the medium containing LE and GL (concentration ratio was 2:3), the conversion rate of GL was 84.12% with a 38.18% increase and the yield of GAMG was 80.47% with an increase of 37.18% than the medium only containing GL at 48h and the enzyme activity of β-glucuronidase was also enhanced from 22.4U/ml to 82.3U/ml. The results showed that the induced expression of PGUS was highly improved when a certain amount of LE was added into the medium.????? | |||

Abstract:Apigenin, being an important dietary compound, is a natural product mainly contained in many vegetables. Growing evidence reveals that the intake of apigenin, which is rich source of celery, is negatively associated with various diseases. However, high-efficient extraction of apigenin, with low cost from complex-component plant vegetable tissue, is still a problem. In this study, the influence of different operational conditions during ultrasonic-assisted beta-glucosidase hydrolysis on apigenin extraction from celery was addressed. Enzymatic hydrolysis showed high apigenin extraction capacity at 0.1 mg/mL concentration, after 15 h, at pH 6. Ultrasonic-assisted enzymatic hydrolysis also achieved high extraction activity towards apigenin with these optimal parameters: an exposure period of 40 min at 60C, power source 60 W, and with 70% methanol concentration | |||

Abstract:In order to know the feasibility of protecting active conformation of enzyme during the covalent-immobilization, three lyophilized enzymes catalase (CAT), trypsin (TRY) and laccase (LAC) are immobilized under organic media and aqueous media, and kinetic constants are investigated. Specific activity of CAT immobilized in chloroform is 1.67 times larger with Kcat 2.43 times higher and Kmapp is 0.74 times smaller than that of CAT immobilized in aqueous phase. Specific activity of TRY immobilized in ethyl acetate is 5.33 times larger with Kcat 55.58 times higher and Kmapp is 0.40 times smaller than that of TRY immobilized in aqueous phase. Specific activity of LAC immobilized in ethyl acetate is 0.24 times larger with Kcat 0.29 times higher and Kmapp is 0.27 times smaller than that of LAC immobilized in aqueous phase.Results show that organic media has more advantages over aqueous media to remarkably improve activities and kinetic constant values. Organic media makes the enzymes under〝rigid〞state, and this "rigid" state helps enzymes maintain their original active conformation and prevent enzymes from conformation disorder during the modification reaction occurring. | |||

Abstract:[Background] Plants have been proven to be promising bioreactors in the production of many heterologous proteins including those in the biopharmaceutical industry. However, the efficiency of a potential plant bioreactor is impaired by the limitation in available promoters to drive foreign gene expression. Hence, the discovery of novel promoters is urgently needed. Seeds are ideal for protein storage and proteins can be kept as a stable form for several months. Thus a strong seed-specific promoter is a powerful tool in directing the expression and accumulation of target proteins in the seed. [Results] A 784-bp 5'-flanking sequence of the mung bean seed storage protein 8S globulin α' subunit gene was isolated by genome walking. When the sequence was analyzed with bioinformatics tools, numerous putative cis-elements were identified. The green fluorescent protein (GFP) reporter driven from this promoter was transiently expressed in protoplasts derived from mung bean cotyledons. Finally, transgenic Arabidopsis plants expressing the β-glucuronidase (GUS) reporter gene driven from the 8S globulin alpha' promoter were generated. GUS expression in transgenic seeds was observed by histochemical and quantitative GUS assays, confirming seed expression and high transcriptional activity. This study has identified the mung bean 8S α' promoter having potential for directing expression in seeds for bioreactor application. | |||
