Abstract:\justifying The endocytosis of 100nm targeted liposomes by endothelium cells in a shear flow is modeled by a set of master equations of the birth and death process. The relations between the shear rate and the parameters in the equations are established to analyze the effect of the shear flow on endocytosis. With the model, we show that the shear-induced variation of the bond formation rate does not influence the endocytosis and the insufficient binding strength of the non-specific attraction between lipid bilayers of liposomes and membranes of cells is the reason for the appreciable decrease in the number of endocytosed liposomes with the shear rate in former experiments. For the endocytosis of liposomes conjugated with PH1 peptide by human umbilical vein endothelial cells which expresses Tie2 receptors on their membranes, it is shown that the shear-elevated expression of Tie2 can evidently increase the number of endocytosed liposomes if the shear rate is larger than $100{\rm s}^{-1}$ and the duration of the experiment is longer than 30min. This result agrees with our experimental data. | |||

Abstract:Intermolecular double-quantum coherence (iDQC) 1H-nuclear magnetic resonance spectroscopy (1H NMR), which serves as a complementary method in the analyses of fatty acids, is used to investigate the intact salmon muscle and the whole zebra fish. The spectra of fatty acids of the intact salmon muscle, whose resonances are overlapped with metabolites peaks in the conventional NMR spectra, can be resolved in the presence of severe intrinsic structural inhomogeneity without sample pretreatments and special NMR accessories such as those for high speed sample spinning. For improving the practicability of the iDQC method, a localized module is combined with the iDQC method so that the fatty acids of the whole zebra fish can be detected non-invasively. All the iDQC results are verified by the extraction NMR spectra. In addition, fatty acid composition of the salmon muscle is quantitatively analyzed based on the iDQC and extraction NMR spectra. The calculated results from these two methods are in good agreement. Therefore, the iDQC method may serve as a feasible one-step and fast screening method for fish quality analyses and lipid inspections of other biological tissues. | |||

Abstract:In this paper, the potential role of CaD in mediating mechanical properties of VSMCs was probed and the regulatory effect of CaD on cell motilities was also discussed. First, structural effect of CaD on cytoskeleton integrity was analyzed with CaD knock-down; Second, Fourier transform traction cytometry was used to measure cell traction force dynamics; Third, the Optical Magnetic Twisting Cytometry was employed to determine cell stiffness dynamics. We found that CaD is an integral component to maintain cytoskeleton integrity of VSMCs. CaD not only sustains the basic value of contraction and stiffness of VSMCs in stationary condition, but also restores these mechanical parameters after cytoskeleton disruption. | |||

Abstract:The functions of IgG mainly rely on the interactions of its Fc region with other proteins. 31 patients with stage IIIB and IV non-small cell lung cancer (NSCLC) were enrolled in this study. The levels of IgG1 Fc N-glycosylation in a series of serum samples from 31 NSCLC patients treated with icotinib hydrochloride were determined by matrix-assisted laser desorption/ionization-Fourier transform ion cyclotron resonance mass spectrometry (MALDI-FTICR MS). Our findings suggest that IgG1 Fc-galactosylation and Fc-agalactosylation levels of NSCLC patients changed with treatment time and gender was associated with changes in the IgG1 Fc- galactosylation and Fc-agalactosylation levels. Gender-specific changes in IgG1 Fc-galactosylation and Fc-agalactosylation may helpful in understanding the mechanism of EGFR drug action. | |||

Abstract:Disease-specific humoral immune response-related protein complexes in blood are associated with disease progression. 31 patients with stage IIIB and IV non-small cell lung cancer (NSCLC) were administered with oral dose of icotinib hydrochloride (150 mg bid or 125 mg tid) for a 28-continuous-day cycle until diseases progressed or unacceptable toxicity occurred. The levels of immunoinflammation-related protein complexes (IIRPCs) in a series of plasma samples from 31 NSCLC patients treated with icotinib hydrochloride were determined by an optimized native-PAGE gel. Our results indicate that the time length of humoral immune and inflammation response (TLHIIR) was closely associated with disease progression, and the median TLHIIR was 22.0 weeks, 95% confidence interval (CI): 16.2 to 33.0 weeks, with a lead time of median 11 weeks related to clinical imaging evidence confirmed by computed tomography or magnetic resonance imaging (the median of progression-free survival, 34.0 weeks, 95% CI: 27.9 to 49.0 weeks). Our findings suggest the existence of a complex relationship between humoral immune response, acquired resistance, and disease progression. Personalized IIRPCs could be an excellent indicator to monitor the disease progression. | |||

Abstract: Nonlinear optical single-molecular image technique is a new technique which is not widely recognized in scientific community. It is a new innovative principle to get a profile image of tiny materials noninvasively. by a series of lenses which diameters from small to large, adjusting each lens to move back or forward carefully along a straight line, the two different direction rays from the same point of the out edge of objects can focus on a plate to form an image. This technique can magnify profile images of small samples. Its x-y resolution breakthrough the limit of Abbe's diffraction law. Nowadays , its resolution can reach 1-5 nanometer . In this paper, we gave more supporting evidences that Nonlinear optical single-molecular image technique is a practical tool. This paper showed the evidences of application in DNA Double helix nanostructure study. The photos of our experimental results demonstrated that the principle of Nonlinear optical single-molecular image technique is correct. This technique will bring human a lot of knowledge and information about molecules, especially about macromolecules in living cell. | |||

Abstract:Red blood cells (RBCs) are very important due to their role of oxygen transport from lungs. As the malaria parasite grows in the malaria-infected red blood cells (IRBCs), the properties of the cells change. In the present work, the oxygen uptake by RBCs and IRBCs at the pulmonary capillaries is simulated using a numerical technique based on the two-dimensional immersed interface method. The results for the oxygen uptake by a stationary single RBC have fair agreements with the previously reported results. The numerical results show that the malaria infection could significantly cause deterioration on the oxygen uptake by red blood cells. The results also suggest that the oxygen uptake by individual stationary RBC/IRBC would not be significantly affected by the neighboring cells provided the separation distance is about the dimension of the cell. ????? | |||

Abstract:Lipases (EC 3.1.1.3) possess two critical conformations, "open" and "close", during their catalytic process. The conformational changes, caused by the flexible loop regions in the enzymes, determine selectivity, regioselectivity and stereoselectivity of substrates in the enzymatic reactions. However, the principles beneath the phenomenon have not been clearly understood thoroughly yet, in terms of mutability on these loop regions. This paper employed both sequence and structure bioinformatical methods to study lipases available in database, especially for loop regions such as the 'lid' domain that was widely reported to connect substrate selectivity. Unsurprisingly, the 'lid' domain is quite mutable in a group of lipases which are in a moderate similarity for sequence and structure. However, we have found a more mutable, even the most mutable, domain among these loop regions. Loop 9, a very close to C terminus, is ranked top 1. The new discovery may rationalize the coupling effect from colipase. | |||

Abstract:Bioluminescent proteins as a high sensitive optical reporters for imaging in living animals, have been widely used in analytical applications in intracellular monitoring, genetic regulation and detection, immune and binding assays. In this work, we have systematically analyzed the information of 199 bioluminescent proteins and nonbioluminescent proteins, and propose an approach for distinguishing bioluminescent proteins by combining the dipeptide composition, reduced amino acid composition, evolutionary information, and auto covariance of averaged chemical shift (acACS). The overall prediction accuracy is 82.16% by Jackknife validation using the algorithm of support vector machine (SVM) and Increment of diversity. This result is better than that obtained by other existing methods. The improvement of the overall prediction accuracy reaches up to 5.33% higher than those of the SVM and auto covariance of sequential evolution information by 10-fold cross-validation. The acACS performance is excellent, indicating that our approach is better than other existing methods in the literature. | |||

Abstract:A method of using Au colloid to capture the decomposed product of penicillin-penicillamine on a surface plasmon resonance (SPR) biosensor for the quantitative determination of penicillin was developed. Based on the de-composition of penicillin to generate penicillamine and penilloaldehyde, a high sensitive biosensor for detecting penicillin was also developed. In our experiment, it was penicillamine rather than penicillin that has been measured. This is because penicillamine contains a functional group that makes it self-assembling on Au colloid to increase the molecular weight so as to improve the surface plasmon resonance signal. On a UV-Vis spectrophotometer, a high concentration of penicilliamine-Au complex was determined, indicating that penicillamine was already well combined with Au colloid. The method, using the combination of Au colloid with penicillamine, proved to detect penicillin. | |||
