Abstract:Exocarpium Citri grandis (ECG, called Huajuhong in Chinese) is traditional Chinese herbal medicine and has been used for the treatment of respiratory diseases for hundreds of years. Contained flavonoids have been documented to be the primary active components. Recently, ECG has been listed in a TCM formula in Guideline for the diagnosis and treatment of COVID-19 (Sixth Edition) in China. However, the efficacy and mechanism of ECG in the prevention and treatment of respiratory diseases have not been systematically reviewed. Hence, In this review, we summarized the efficacies of ECG and its pharmacologically active compound naringin in antitussive, expectorant, improving lung function, alleviating acute lung injury, attenuating pulmonary fibrosis, and enhancing antiviral immune response, so as to provide a reference for its clinical application. | |||

Abstract:Fructose excess consumption induces insulin resistance-associated metabolic diseases. Hypothalamic insulin signaling plays a pivotal role in controlling whole-body insulin sensitivity and energy homeostasis. Wuling San and its modifications exerted various effects including anti-hypertension, anti-inflammation, anti-dyslipidemia and anti-insulin resistance. In this study, we investigated its regulatory effects on the hypothalamus of high fructose-fed mice. Mice were fed 30% fructose in drinking water for 12 weeks. After 6 weeks, these animals were orally treated with Wuling San (987, 1316, 1755 and 2340 mg/kg), allopurinol (5 mg/kg) and water daily for the next 6 weeks, respectively. Wuling San ameliorated fructose-induced hypertension, hyperlipidemia, and hyperinsulinemia and insulin resistance in mice. Wuling San ameliorated high fructose-caused hypothalamic inflammation in mice by suppressing the activation of hypothalamic nuclear factor κB (NF-κB) pathway and NOD-like receptor 3 (NLRP3) inflammasome. Wuling San also effectively restored high fructose-induced hypothalamic insulin signaling defect by up-regulating the phosphorylation of insulin receptor and protein kinase B. Allopurinol had similar effects. These results provide in vivo evidence that Wuling San-mediated inhibition of NF-κB pathway/NLRP3 inflammasome activation in the hypothalamus of mice may be associated with the reduction of hypothalamic inflammatory lesions, contributing to the improvement of hypothalamic insulin signaling defect in this model. Thus, Wuling San with the central activity may be a therapeutic for high fructose-induced metabolic syndrome in humans. | |||

Abstract:A simple, sensitive, and accurate LC-MS/MS method for determining triptolide in mouse plasma has been developed and validated to study the drug-drug interaction with curcumin. The quantification was performed with the transitions of [M+NH4]+→[M+H]+ for triptolide. The assay for triptolide in mouse plasma was linear over the range of 0.25 to 100 ng/mL and the Lower Limit of Quantification was 0.25 ng/ml. After a single oral administration of triptolide at 0.5 mg/kg, triptolide reached the maximum exposure at 11.64 ± 4.08 min, and declined rapidly with a mean elimination half-life of 38.52 min. Co-administration of 400 mg/kg curcumin significantly increased Cmax and AUC0-∞ by 1.8-fold and 1.5-fold respectively, demonstrating that curcumin increases the bioavailability of triptolide in mouse. | |||

Abstract:The stem of Marsdenia tenacissima (Roxb.) Wight et Arn. has long been used as a medicine to treat cancer in China. Our previous results showed that Marsdenia tenacissima extract (MTE) overcomes gefitinib resistance in NSCLC cells. The present study investigated in vivo anti-tumor activity of MTE combined with gefitinib. H460 (K-ras mutation) or H1975 cells (T790M mutation) was subcutaneously inoculated into nude mice. Tumor volume and body weight were measured during the experiment. The resected tumors were weighed after animals were sacrificed. Cellular proliferation and apoptosis in xenografts tumor tissue were assessed. EGFR downstream pathways and c-Met expression was evaluated by western blotting. In accordance with the previous in vitro study, MTE at low dose (5 g/kg) was chosen to assess whether it can restore gefitinib sensitivity in vivo. Interestingly, MTE prominently enhanced gefitinib efficacy in the resistant H460 and H1975 xenografts. This combination significantly inhibited tumor proliferation and induced cell apoptosis in both resistant NSCLC xenografts. Constitutive activation of PI3K/Akt and MEK/ERK pathway is related to EGFR-TKI resistance. Accordingly, phosphorylation of PI3K/Akt/mTOR and ERK1/2 was suppressed after the combined treatment. Simultaneously, the cross-talked c-Met and EGFR was also prominently lowered in the presence of MTE combined with gefitinib. This study provides in vivo evidence to demonstrate that MTE enhancing gefitinib efficacy in the resistant NSCLC xenografts, and suggests the combination of MTE and gefitinib as a promising approach against NSCLC. | |||

Abstract:Traditional Chinese medicine (TCM) for activating blood and resolving stasis (Huoxuehuayu) has been widely applied in treatment of various platelet-related diseases. The present study was to explore the antiplatelet activity of the water extracts from six kinds of Huoxuehuayu herbs commonly used in clinical context, which fall into three categories, namely Hexue, Huoxue, and Poxue. We also investigated the differences in the antiplatelet activity of the water extracts from three categories. To this end, human platelet aggregation assay induced by ADP and thrombin was used to evaluate the antiplatelet activity, and flow cytometry assay for detecting the effects on the expression of CD41 and CD62P, two activated platelet surface markers. Results showed that all the water extracts (1 mg/ml) could significantly suppress human platelet aggregation in vitro (P<0.01), but extracts from three categories exhibited different inhibitory effects on expression of CD41 and CD62P respectively, suggesting the different properties for Huoxuehuayu. We concluded that combined use of Huoxuehuayu herbs may have potent therapeutic effects against platelet-related diseases in clinical contexts. | |||

Abstract:To investigate the mechanism of suppressive effect of Chinese medicine on Coronary Atherosclerotic Heart Disease (IHD) by observing the effect of Salvia miltiorrhiza injection coordinated with Acanthopanax senticosus injection(MSAI)on the acute myocardial ischemia rat. The optimal formula of MSAI was ascertained using design by uniform distribution. From pharmacodynamics, the rat was applied with isolated heart perfusion method, for measuring Coronary Artery Fluence . Further, Coronary Artery in rat was received physic liquor by intravenous injection using constant flow pump to determine left ventricular systolic pressure (LVSP) , left ventricular maximum pressure variation rate(LVMPVR)and left ventricular end-diastolic pressure(LVEDP). The MSAI was found that could significantly increase CF of the isolated heart of the rats(P<0.01),compared with SI and AI control group, respectively ; in addition,the effect of Formula (Ⅲ)(syringoside : protocatechuic aldehyde = 1:1.25)is the optimization. The results of hemodynamic effect of acute myocardial ischemia in 15min rat treated with MSAI demonstrated that MSAI could remarkably raise LVSP, LVMPVR(±dp/dtmax)and lower LVEDP. Additionally, the effect on resisting acute-myocardial-ischemia of MSAI group was better than that of SI group and AI group(P<0.01). The mechanism of suppressive effect of MSAI on IHD of may be realized by increasing CF, elevating LVSP, LVMPVR(±dp/dtmax),and lowering LVEDP. | |||

Abstract:The aim of the present study was to evaluate the diuretic effect on the urinary electrolyte concentration(Na+, K+, Cl-) and regulation of the relative mRNA expression of aquaporin-1(AQP1), aquaporin-2(AQP2), aquaporin-3(AQP3) and vasopressin V2 receptor (V2R)post oral administration of sclerotia of P.umbellata aqueous extract in normal rats. Aqueous extract of sclerotia of Polyporus umbellatus (50mg/kg, 250mg/kg, 500mg/kg) or the reference drug, furosemide (10mg/kg) were administrated orally to male SD rats and their urine output was quantified and collected 24h and 8days after the treatment. The kidney medulla AQP1, AQP2, AQP3 and V2R mRNA relative expressions were measured with RT-PCR. After single dose of the exact of sclerotia of P.umbellata, urine output was found to be significantly increased, which began at 4h, and at 24h after the treatment, the sclerotia of Polyporus umbellatus extract and furosemide treatment produced the similar total volume of urine excreted. The extract increase urinary levels of Na+, K+, Cl-, to about the same extent, while furosemide increased urinary levels of Na+ and Cl-. After the 8-day doses, all two substances induced significant diuresis, natriuresis and chloriuresis. These two substances don’t regulate the AQP1 and AQP3 mRNA level in normal rat kidney medulla. The AQP2 mRNA level of sclerotia of P.umbellata extract was down-regulated significantly, the V2R mRNA level of sclerotia of P.umbellata extract 50mg/kg dose group and 250mg/kg dose group were down-regulated significantly too. Interestingly, the low-dose group had higher effect on regulation of AQP2 and V2R mRNA level. Aqueous extract of sclerotia of Polyporus umbellatus has conspicuous diuretic effect confirming its ethnopharmacological use. From the pattern of excretion of water, sodium, potassium, cholorine, AQP2 and V2R mRNA level, it may be logically concluded that it has effect from down-regulating AQP2, and down-regulate AQP2 by down-regulating V2R. | |||
