Abstract:Cytochrome P450 mono-oxygenases (P450s) are a superfamily of heme-dependent enzymes which play important roles in almost all of organisms. Based on rice genome database, a gene encoding a rice cytochrome P450 mono-oxygenase was cloned and sequenced (GenBank accession no. AY579214). The coding region of the gene was 1530 nucleotides in length and encoding a protein with 510 amino acid residues in length. Sequence analyses indicated that the cytochrome P450 corresponds to an ent-kaurene oxidase (KO) like protein, which is likely to be involved in phytoanticipins/phytoalexins biosynthesis. Southern blots analysis suggested that there are at least four similar genes encoding ent-kaurene oxidase or ent-kaurene oxidase like proteins in rice genome. Therefore, the gene reported in this paper was named OsKOS1. RT-PCR analysis showed that OsKOS1 was expressed in vegetative organs and reproductive organs, but at a higher level in vegetative organs. The expression level of OsKOS1 in RDV (rice dwarf virus)-infected plants was much lower than that in healthy plants, indicating that OsKOS1 is likely to be involved in defending RDV. | |||

Abstract:eeds of Triticum aestivum L. cv. Zhengyin No1 were treated by 10 μmol/L GA3 and 10 μmol /L ABA before germination respectively. The growth of root and shoot, contents of endosperm amylose and soluble carbohydrate, medial pH value, the utilizing efficiency of storage materials (MUE), and activity of α-amylase in endosperm during seed germination under osmotic stress were investigated. The results showed that shoot growth was promoted by GA3 and inhibited by ABA and osmotic stress. Root growth was promoted by GA3, and retarded by osmotic stress and ABA at early stage. But the growth of root of seedlings from seeds treated by ABA was promoted significantly during the late germination stage. Seedling growth was positively correlated with medial pH and MUE significantly. GA3 promoted the expression and increase in activity of α-amylase in endosperm of wheat seed, resulting in increase in breakdown of amylose to soluble carbohydrates, but ABA and osmotic stress had the opposite effect to GA | |||

Abstract:Effects of silver nitrate on callus regeneration and activity of indole-3-acetic acid oxidase and peroxidase, glutathione S-transferase and reductase, aldehyde oxidase, catalase and peroxidase in cells of immature inflorescence of Triticum aestivum L. variety JI135 were investigated. Immature inflorescences with length between 5-10mm were treated with AgNO3 in MS medium in a period of 10 days. The results indicated that callus regeneration was improved by treatment of wheat immature inflorescence with 16 mg/L AgNO3 for 7 days. The activity of indole-3-acetic acid oxidase and peroxidase was reduced obviously by 16 mg/L AgNO3 four days and six days after inoculation respectively, activity of glutathione transferase, glutathione reductase,catalase,and peroxidase were obviously promoted by AgNO3 4 days after the treatment, But changes in activity of the aldehyde oxidase was not significantly influenced by AgNO3. The mechanism of AgNO3 in improving cell regeneration was discussed. | |||

Abstract:e did the same experiment as Lewis’ (1994), but came out with different kinetics of embolus collapse in the tracheids of stem segments of Platycladus Orientalis L.. Tangential wood sections were trimmed. They were observed under a microscope, while segments were dehydrated and rehydrated. As in Lewis’ experiment, following the initial contract of the gas in the tracheids of a segment, the elongated bubbles began to shrink. Then, we found that there were spherical bubbles between coverslip and glass slide, and the bigger ones were getting larger and the smaller ones contracted gradually while the elongated bubbles in the tracheids collapsed. By calculation, the total amount of gas in all bubbles at the beginning of the collapse of the elongated bubbles was almost the same as the amount of gas midway and at end.
