Abstract:Fruit mature and carotene biosynthesis is adversely affected by high temperature. The objective of this study was to understand the respective impact of exogenous spermidine on ripening, fruit quality and carotene content with off-vine tomato under high temperature. Mature green tomato fruit (Lycopersicon esculentum Mill) of cv. 'zhongshu 6' were pretreated with spermidine solution and then kept under different temperature regimes. The color, firmness and composition changes were examined during storage. Exogenous spermidine and high temperature treatment can severely affect the ripening process and the final nutrient composition of tomato fruit. Under normal temperature, exogenous spermidine enhanced fruit TCI value, total carotene content and ethylene production. Increasing temperature from 26 to 32 ℃, reduced lycopene content, but increased ethylene production. Exogenous application of spermidine could improve the heat resistance and partially alleviate the affect of high temperature, which might attribute to the higher endogenous level of Put and Spm.

Abstract:The kinetic parameters of partially purified phenoloxidase (PO, EC. 1.14.18.1) from the larvae of Anomala corpulenta M. were determined, using L-3, 4-dihydroxyphenylalanine (L-DOPA) as substrate. The optimal pH and temperature of the enzyme for the oxidation of L-DOPA were determined to be at pH 7.5 and at 50 C, respectively. At pH 7.5 and 50 C, the Michaelis constant (Km) and maximal velocity (Vm) of the enzyme for the oxidation of L-DOPA were determined to be 11.11 mmol/L and 107.53 U/mg, respectively, and the Km and Vm of PO for the oxidation of catechol were 9.52 mmol/L and 41.67 U/mg, respectively. Morin could effectively inhibit the activity of phenoloxidase and this inhibition was reversible and competitive, with the IC50 of 1.05 mmol/L. The inhibition constants were estimated to be 0.54 mmol/L.
Abstract:Cytoplasmic male sterility (CMS) is not only important for exploiting heterosis in crop plants, but also as a model for investigating nuclear-cytoplasmic interaction. CMS may be caused by mutations, rearrangement, or recombination in the mitochondrial genome. With the use of the next-generation sequencing technology, we sequenced the mitochondrial genome of hau CMS line and its maintainer line in B. juneca. The result showed that B. juncea hau CMS mitochondrial genome was assembled into a single, circular-mapping molecule with a size of 247,903 bp with 45.08% GC content. The genome contains 35 proteins, 3 rRNAs, 25 tRNA genes and 29 ORFs of unknown function in addition to the CMS associated gene orf288. The sizes of the maintainer line and another normal type line "J163-4" are both 219,863 bp and the GC content is 45.23%. The maintainer line contains 36 proteins, 3 rRNAs, and 22 tRNA genes and 31 unidentified ORFs. After the comparative analysis the hau CMS line and its maintainer line, specific markers were developed based on their unique ORFs. These can be used as an effective markers to separate the hau CMS line from its maintainer line at the seedling stage. We also confirmed that substoichiometrically different mitotypes coexist in hau CMS lines and its maintainer lines in B. juncea. The number of repeats larger than 100bp in the hau CMS line (16 repeats) are almost twice those of the maintainer line (9 repeats). The 5' region of the chimeric gene orf288 is highly similar to nad5, and the 3' region is also similar to another predicted mitochondrial ORF-orf293 in B. juncea. Phylogenetic analysis of the CMS-associated gene orf288 and four other homologous sequences in Brassicaceae showed that orf288 bore strong similarity to orf263 in Brassica tournefortii, but they still differed from one another. The hau CMS mitochondrial genome was highly rearranged when compared with its iso-nuclear maintainer line mitochondrial genome as the other CMS lines in different crops. The gene orf288 was composed of part nad5, orf293 and an unknown origin in the 5' region as orf263 reported in Brassica tournefortii. It may help us identify the mechanism for natural CMS in B. juncea, and compared with the sequenced mitochondrial genomes in Brassicas, uncover the origin of the hau CMS mitotype and structural and evolutionary differences between different mitotypes.

Abstract:Orientational planting cannot only make maize leaf growing consistently, but also improve the leaf photosynthetic capacity of per unit area and yield of maize. The "tip" direction was detected and the angle of deflection was measured by using contour curvature analysis method of maize seed, the S component of HSV space was preprocessed by Otsu method, the automatic extracting maize seeds embryo and orientation information have been realized through image color space conversion, segmentation, preprocessing, and its contour characteristic analysis. A rectangular region ROI in the image was defined and counts up pixels within the region, the region-specific positive and negative ROI pixels were compared by TM threshold and the embryo side towards was identified. The Zheng-dan958, Jun-dan20 and Zhong-ke11, and repeat three times by using the above methods. The results showed that the average accuracy of embryo inspection was more than 95%; the direction average angle was 2.2 .
Abstract:A common leafhopper pest on peach trees, Táoyīdiǎnyèchán (桃一点叶蝉), has previously been misidentified as Erythroneura sudra (Distant, 1908). The correct identification is here shown to be Singapora shinshana (Matsumura, 1932) based on available descriptions, illustrations and records of distribution and host plants. Habitus photos and illustrations of the male genitalia for both species are provided for comparison.

Abstract:Self-incompatibility (SI) is one important factor that can result in seedless fruits of Citrus. Our previous study showed that 'Wuzishatangju' mandarin was gametophytic self-incompatibility (GSI) and 72 h was the crucial stage for the shift from a self-compatible to a self-incompatible state. However, it is not clear what genes are involved in the process. In this study, two SSH libraries were constructed to screen differentially expressed genes using 72 h styles after self-pollination of 'Wuzishatangju' and cross-pollination of 'Wuzishatangju' × 'Shatangju'. 106 and 97 differentially expressed cDNA clones were sequenced and identified from forward and reverse SSH libraries, respectively. Differentially expressed ESTs are possibly involved in the SI reaction of 'Wuzishatangju' through S-phase kinase-associated protein1 (SKP1)-like activity, autoinhibited Ca2+-ATPase 1, U-box domain protein, and serine/threonine kinase. Results from quantitative real-time PCR (qPCR) showed that SKP1-like and U-box were obviously up-regulated in ovaries before pollination of 'Wuzishatangju', and approximately 20-and 2-fold higher than that of 'Shatangju', respectively. The SKP1-like and U-box were obviously up-regulated in pistils at 4 d after self-pollination of 'Wuzishatangju', and approximately 20-and 800-fold higher than that the same stage after cross-pollination of 'Wuzishatangju' × 'Shatangju', respectively. The potential involvement of these genes in the SI reaction is discussed.

Abstract:It detected the canopy temperature of rice via automated infrared imaging technology in the test under different irrigation condition, and used the CWSI theoretical model to diagnose whether the crop suffered water stress or not. It also analyzed the water stress index theoretical model of crop and other indexes on reflecting the water status of crop, including the relationship between the CWSI and leaf stomatal resistance, the CWSI and leaf net photosynthetic rate, and the CWSI and the soil moisture content. The results showed that the relations between the surface theoretical model and the above indexes were fine. It meant the CWSI well reflected the features of water stress of rice.

Abstract:Apoptosis is executed through the activity of the caspases that are aspartyl-specific proteases. In this study, we isolated the caspase gene (Cscaspase-1) of Chilo suppressalis (one of the leading pests responsible for destruction of rice crops). It possesses the open reading frame (ORF) of 295 amino acids including prodomain, large subunit and small subunits, and two cleavage sites (Asp23 and Asp194) were found to be located among them. In addition to these profiles, Cscaspase-1 contains two active sites (His134 and Cys176). Genomic analysis demonstrated there was no intron in the genome of Cscaspase-1. The Cscaspase-1 transcripts were found in all tissues of the fifth instar larvae, and higher levels were found in the midgut, hindgut and Malpighian tubules. Examination of Cscaspase-1 expression in different developmental stages indicated low constitutive levels in the eggs and early larvae stages, and higher abundances were exhibited in the last larvae and pupae stages. The relative mRNA levels of Cscaspase-1 were induced by heat and cold temperatures. For example, the highest increase of Cscaspase-1 transcription was at ?3 C and 36 C respectively. In a word, Cscaspase-1 plays a role of effector in the apoptosis of C. suppressalis. It also correlates with development, metamorphosis and thermotolerance of C. suppreassalis.

Abstract:Observed by infrared thermal imaging technology, in the test rice canopy temperature were measured under different irrigation conditions, taking the Idso empirical model to determine the lower bound equation of the canopy-air temperature differences, and so to get the best period of observing the rice water deficit by the CWSI, in addition the relationship of the CWSI, soil water content and transpiration rate were also analyzed. The preliminary test analysis showed that the CWSI can reflect the rice water stress conditions well.

Abstract:Effect of Oolong tea polysaccharide (OTPS) on the antioxidant and antidiabetic activities in streptozotocin-induced diabetic rats were investigated. Significant changes of biochemical were found in serum and tissues 30 days after OPTS-treated diabetic rats induced by a single dose of streptozotocin. OTPS significantly reduced blood glucose (BG), nitric oxide (NO) and malondialdehyde (MDA) concentration and activity of nitric oxide synthase (NOS) in serum of diabetic rats, meanwhile the activities of superoxide dismutase(SOD), glutathione peroxidase (GSH-PX), and the content of insulin in serum were increased.
